Asymmetric Schiff base manganese(III) complexes involving salen ligands, N,N’-bis(salicylidene)2,3-diaminopyridine, N,N’-bis(3-methoxysalicylidene)2,3-diaminopyridine, N,N’-bis(3,5-di-tert-butylsalicylidene)2,3-diaminopyridine and N,N’-bis(3,5-di-chloro-salicylidene)2,3-diaminopyridine were prepared and their catalytic activity was investigated in the oxidation of some primary and secondary alcohols. During optimization of oxidation reactions, Mn-4, bearing electron withdrawing N,N’-Bis(3,5-di-chloro-salicylidene)2,3-diaminopyridine ligand, showed higher activity than other catalysts tested. The catalytic reactions were carried out in the presence of various oxidants such as oxygen, hydrogen peroxide or tert-butyl hydroperoxide (TBHP) and additives such as acetic acid and imidazole. The oxidant/additive combination of TBHP and imidazole was shown to be effective for the oxidation process and the degree of their impact on oxidation reaction was found highly dependent on a balanced ratio between them. Mn-4 was selected as the most effective catalyst under optimized reaction conditions and revealed efficient for the oxidation of secondary alcohols.

Searching for CNS active cyclic amines derivatives containing heterocyclic xanthone core we designed and synthesized a set of fourteen novel 2- or 4-methylxanthone substituted by alkyl- or aryl-piperazine moieties. The compounds were evaluated in vivo for their potential antidepressant-like activity (in the forced swim test) and anxiolytic-like activity (four-plate test) and their inhibitory effect against rat 5-HT2 receptor was checked. The pharmacokinetic analysis of active compounds done by a non-compartmental approach have shown a rapid absorption of all studied molecules from intraperitoneal cavity and good penetration the blood-brain barrier after i.p. administration with brain to plasma ratios varied from 2.8 to 31.6. Genotoxicity and biotransformation of active compounds were studied. Compound 19 interactions with major classes of GPCRs, uptake systems and ion channels were tested and results indicated that it binds to 5-HT2A, 5-HT2B receptors and sodium channels.

Four Schiff base manganese(III) complexes with derivatives of [(R,R)-N,N’-bis(salicy1idene)-1,2-cyclohexanediaminato)] including substituents on salicylaldehyde such as 3-methoxy, 3,5-di-tert-butyl and 3,5-chloro were synthesized and characterized using a combination of IR, UV-Vis, and HR ESI-MS techniques. The catalytic activity of these complexes was tested in the oxidation of 1-phenylethanol to acetophenone, revealing very good performances for all of the four manganese complexes. The catalytic reactions were carried out in the presence of tert-butyl hydroperoxide (TBHP) as oxidant and imidazole as co-catalyst. Complex Mn-4, bearing electron withdrawing [(R,R)-N,N’-bis(3,5-di-chloro-salicylidene)-1,2-cyclohexanediaminato)] ligand was found to be the most stable of the tested Mn(III) complexes and was selected for the oxidation of several primary and secondary alcohols. A novel third-nonlinear optical (NLO) crystal, piperazine-1,4-diium bis(2,4-dichlorobenzoate) (abbreviated as PBDCB) was successfully synthesized and grown good quality crystal by slow evaporation solution growth technique by methanol as a solvent at room temperature. The formation of the grown crystal was carried out by single-crystal X-ray diffraction analysis. The analysis of a single crystal X-ray diffraction reveals that the title compound was crystallized in the monoclinic system with centrosymmetric space group P21/c and thereof lattice parameter are a = 11.166(6) angstrom, b = 8.160(5) angstrom, c = 12.085(6) angstrom, beta= 113.18(3) degrees, Z = 4. Crystallographic studies also reveal that the molecules are linked via N-HO and C-H Cl hydrogen bonds. Pairs of intermolecular N-HO hydrogen bonds generate R 4 4 (12) ring motifs. The powder X-ray diffraction study was completed and the peaks were indexed. UV-Vis spectral studies were revealed that the grown crystal is transparent in the entire visible region with a lower cut off the wavelength of 286 nm. The optical band gap of the title compound was measured by Tauc’s plot method, and it was obtained to be 3.9 eV, which shows suitable applications of the optoelectronic device. In the photoluminescence study, the grown crystal was revealed violet emission radiation, and thereof peak has appeared at 312 nm. The functional groups of the grown crystal have been determined by Fourier Transform Infra-Red (FTIR) spectroscopy technique. In thermogravimetric (TGA) and differential thermal analysis (DTA) analysis, the title material was determined as thermally as stable up to 163 degrees C. The Vickers Microhardness test ascertained the mechanical properties of the grown crystal material, and it belongs to a soft material category. The dielectric measurements of the developed crystal material were also ascertained at various frequencies. The dielectric constant, dielectric loss, and A.C. the conductivity of title material was studied with different frequencies and temperatures. The laser damage threshold (LDT) of the grown crystal was then calculated by using Nd: YAG laser. The grown crystal has given the measured (LDT) value of 8.97 GW/cm2. The third-order nonlinear optical (NLO) properties characteristics, such as absorption coefficient (beta similar to 0.02 x 10 -4 cm/W), the refractive index (n(2) similar to 4.06 x 10(-8) cm(2)/W), and their susceptibility range values of (chi((3)) similar to 2.17 x 10(-6)esu) of the grown crystal was estimated by Z-scan technique, suggests that the grown crystal material PBDCB was served as a promising source for nonlinear optical devices. The insect enzyme GH20 beta-N-acetyl-D-hexosaminidase OfHex1 represents an important chitinolytic enzyme found in the agricultural pest Ostrinia furnacalis (Guenee) and inhibition of this enzyme has been considered a promising strategy for the development of eco-friendly pesticides. In this article, based on the structure of the catalytic domains of OfHex1, a series of novel glycosyl triazoles were designed and synthesized via Cu-catalyzed azide-alkyne [3+ 2] cycloaddition reaction. To investigate the potency and selectivity of these glycosyl triazoles, the inhibition activities towards OfHex1 and HsHexB (human beta-N-acetylhexosaminidase B) were studied. Particularly compound 17c (OfHex1, K-i= 28.68 mu M; HsHexB, K-i > 100 mu M) exhibited a suitable activity and selectivity against OfHex1. Furthermore, the possible inhibitory mechanisms of 17c with OfHex1 were studied using molecular docking and MD simulations. The structure-activity relationship results as well as the formed binding patterns may provide promising insights into the further development of novel OfHex1 inhibitors.

We report here the stereoselective bioreduction of -nitro ketones catalyzed by ketoreductases (KREDs) with publicly known sequences. YGL039w and RasADH/SyADH were able to reduce 23 class I substrates (1-aryl-2-nitro-1-ethanone (1)) and ten class II substrates (1-aryloxy-3-nitro-2-propanone (4)) to furnish both enantiomers of the corresponding -nitro alcohols, with good-to-excellent conversions (up to >99%) and enantioselectivities (up to >99% ee) being achieved in most cases. To the best of our knowledge, KRED-mediated reduction of class II -nitro ketones (1-aryloxy-3-nitro-2-propanone (4)) is unprecedented. Select -nitro alcohols, including the synthetic intermediates of bioactive molecules (R)-tembamide, (S)-tembamide, (S)-moprolol, (S)-toliprolol and (S)-propanolol, were stereoselectively synthesized in preparative scale with 42% to 90% isolated yields, showcasing the practical potential of our developed system in organic synthesis. Finally, the advantage of using KREDs with known sequence was demonstrated by whole-cell catalysis, in which -nitro alcohol (R)-2k, the key synthetic intermediate of hypoglycemic natural product (R)-tembamide, was produced in a space-time yield of 178 g L-1 d(-1) as well as 95% ee by employing the whole cells of a recombinant E. coli strain coexpressing RasADH and glucose dehydrogenase as the biocatalyst.

As our research focuses on anticancer drugs, a series of novel derivatives of flexicaulin A (FA), an ent-kaurene diterpene, condensed with an aromatic ring were synthesized, and their antiproliferative activities against four human cancer cell lines (TE-1, EC109, MCF-7, and MGC-803) were evaluated. The activities of most of the new compounds were better than those of FA. Compound 2y exhibited the best activity with an IC50 value reaching 0.13 mu M against oesophageal cancer cells (EC109 cells). The IC50 values for 2y in normal cells (GES-1 cells and HUVECs) were 0.52 mu M and 0.49 mu M, respectively. Subsequent mechanistic investigations found that compound 2y can inhibit the proliferation of cancer cells and cell cloning. In addition, 2y could reduce the mitochondrial membrane potential, increase the apoptosis rate, and increase the ROS level in EC109 cells. Moreover, 2y can upregulate the expression of ROS/JNK pathway-related proteins (p-ASK1, p-MKK4, p-JNK, and p-Cjun (ser63)) and pro-apoptotic proteins (Bax, Bad, and Bim). In vivo experiments showed that 2y can inhibit tumour growth in nude mice. The mechanism involves an increase in protein expression in the ROS pathway, leading to changes in apoptosis-related proteins. In addition, compound 2y shows low toxicity. Oxathiapiprolin is one of the best active fungicides discovered for oomycetes control. To develop a fungicide candidate with a broad spectrum of activity, 22 new piperidinylthiazole derivatives were designed and synthesized. Compound 51 showed the best activity against Pseudoperonospora cubensis (Berk. et Curt.) Rostov and Phytophthora infestans in vivo with 100% and 80% of inhibition, respectively, at 1 mg/L, and 72.87% of field efficacy against P. cubensis at 1 g ai/667 m(2) validated these results. Compound 5i exhibited a broad spectrum of excellent activity against Sclerotinia sclerotiorum with EC50 = 0.30 mg/ L (>10 times more active than oxathiapiprolin and azoxystrobin in vitro), good activity against Botrytis cinerea, Cercospora arachidicola, and Gibberella zeae with EC50 of 14.54, 5.57, and 14.03 mg/L in vitro and against P. cubensis and P. infestans with 60% and 30% inhibition rates, respectively, at 1 mg/L in vivo. Mode of action studies by RNA sequencing analysis discovered oxysterol-binding protein (OSBP), chitin synthase (CHS1), and (1,3)-beta-glucan synthase (FKS2) as the potent target of 5i against S. sclerotiorum. Quenching studies validated that OSBP was the same target of both 5i and oxathiapiprolin; it was quenched by both of them. Our studies discovered isothiazole-containing piperidinylthiazole as an OSBP target-based novel lead for fungicide development.

Three kinds of novel hydroxyapatite@terbium complex core-shell composites were synthesized with 2-chlorobenzoic acid, 4-chlorobenzoic acid, and 2,4-dichlorobenzoic acid as the ligand, respectively. The XRD, UV/vis absorption and FT-IR results show that terbium ions are coordinated with ligands and the corresponding complexes are successfully included into composites. SEM and EDS results reveal that Tb complexes are fully and homogenously coated on the surface of hydroxyapatite (HAP) microspheres consisting of a large number of nanosized crystals. The strongest luminescence properties are obtained when 2,4-dichlorobenzoic acid is used as ligand. The study on fluorescence lifetime of composites shows that the lifetime is inversely related to emission intensity. TGA-DSC results indicate that the core-shell composites have an excellent thermal stability. Therefore, the current core-shell structure provides a cost-effective route to manufacture of biocompatible and heat-resistant highly luminescent materials. (C) 2020 Chinese Society of Rare Earths. Four Schiff base manganese(III) complexes with derivatives of [(R,R)-N,N’-bis(salicy1idene)-1,2-cyclohexanediaminato)] including substituents on salicylaldehyde such as 3-methoxy, 3,5-di-tert-butyl and 3,5-chloro were synthesized and characterized using a combination of IR, UV-Vis, and HR ESI-MS techniques. The catalytic activity of these complexes was tested in the oxidation of 1-phenylethanol to acetophenone, revealing very good performances for all of the four manganese complexes. The catalytic reactions were carried out in the presence of tert-butyl hydroperoxide (TBHP) as oxidant and imidazole as co-catalyst. Complex Mn-4, bearing electron withdrawing [(R,R)-N,N’-bis(3,5-di-chloro-salicylidene)-1,2-cyclohexanediaminato)] ligand was found to be the most stable of the tested Mn(III) complexes and was selected for the oxidation of several primary and secondary alcohols.
