Benzothiazole-based pyrazole derivatives were prepared by a mol. hybridization approach with an aim to influence the biol. activity significantly. Efficient conversion of hydrazones I (R = H, OMe, Cl, etc.; R’ = H, F; R” = H, Me, Br, Cl) derived from their corresponding Me ketones to 4-formylpyrazoles II was carried out at 60° in dioxane, using the Vilsmeier-Haack reagent isolated from phthaloyl dichloride and N,N-dimethylformamide. The newly synthesized compounds II and hydrazones I were screened in vitro for their antimicrobial activities using the broth macrodilution method. The compounds I were also screened for their antioxidant activities using the DPPH radical scavenging assay. Some of the synthesized compounds showed significant antibacterial and antifungal activities as evident from their min. inhibitory concentrations Compound II (R = R’ = H; R” = Br) showed remarkable antioxidant activity.

The derivatives of I exist in two isomeric forms at room temperature in DMSO-d6: (2E,5Z,7E) (2Z,5Z,7E). The stereodynamics of the synthesized derivatives I were investigated using variable temperature dynamic 1H-NMR (VT DNMR). The ΔGâ€?/sup> values (â‰?5 kcal mol-1) estimated for the dynamic process depicted a significant barrier between two forms in solution at ambient temperature To go a step further, line shape anal. were also performed to get a clear understanding of the equilibration mechanism.

A simple method for the one-pot reaction of 3-acetyl-2H-chromen-2-one with different heterylthiols and phenylthioureas under green conditions using tetrabutylammonium tribromide (TBATB) as an efficient reagent has been described. 3-Acetyl-2H-chromen-2-one was reacted with TBATB in acetic acid to give 3-(2-bromoacetyl)-2H-chromen-2-one. Poly(ethylene glycol) 600 (PEG-600) was found to be the effective solvent for this reaction. Thus, a one-pot reaction of equimolar amounts of 3-acetyl-2H-chromen-2-one, TBATB and heterylthiols or phenylthioureas, resulted in the formation of heterylthioacetylcoumarins I (X = NH, O, S) or coumarinylthiazoles II (Ar = C6H5, 4-FC6H4, 2-Cl-4-FC6H3, etc.), resp. The effect of the solvent on these reactions was studied. The merits of this preparation are the mild reaction conditions, easy workup, good yields and use of PEG-600 as a green reaction medium.

The synthesis of polymer supported zinc-salen complex (PS-Zn-salen) is described. The mononuclear zinc(II)-salen complex was characterized by Fourier-transform NMR spectroscopy, energy-dispersive X-ray spectroscopy, Fourier transform IR spectrophotometry (FT-IR), thermogravimetric anal., SEM, surface area and pore size distribution by Brunauer-Emmett-Teller. The synthesized PS-Zn-salen complex used as a recyclable heterogeneous catalyst for the efficient synthesis of hydantoins, thiohydantoins and Schiff bases in an aqueous medium. The isolated yields of hydantoins, thiohydantoins and Schiff bases achieved up to 89, 95 and 94%, resp. In spite of conventional heterogeneous catalysts, current PS-Zn-salen complex shows thermal stability up to 280 °C. Moreover, the catalyst could be recovered easily by simple filtration and reused for next run with slightly declining its activity up to six successive runs. The FT-IR spectrum of recycle catalyst after 6th run confirmed that the catalyst was stable during the course of a reaction. The leaching of metal from the PS-Zn-salen is negligible, which was confirmed by AAS and hot filtration test.

In-vivo test results showed that the compounds I [R1 = 4-FC6H4, 4-ClC6H4, 3-Cl-4-FC6H3, 2,4-Cl2C6H3, 4-BrC6H4] exhibited good anti-inflammatory and analgesic activities, similar to that of indomethacin and aspirin, resp. Compounds II showed better anti-oxidant activity than compounds I when compared to ascorbic acid. Further, a mol. docking study was performed to predict the possible binding modes on cyclooxygenase-1 (COX-1) and COX-2 for compounds I and II. Good correlation was observed between the anti-inflammatory activity of the compounds I and II and the results of the binding modes in COX-2.

The present work describes the synthesis of alkyl substituted N,4-di-Ph thiazole-2-amines (I) and (II) through a three step pathway. Further, the structures were confirmed by single crystal X-ray diffraction studies which revealed that compound I crystallizes in triclinic system with P-1 space group whereas compound II crystallizes as monoclinic with P21/c space group. The asym. unit consists of only one mol. in both the structures. The study of torsional angles of compound I and compound II indicated that S1 and C4 are cis to each other whereas N2 and C4 are trans. A detailed anal. of intermol. hydrogen bonding interactions has been performed. Also their C-H···π and π···π stacking interactions have been explored using Hirshfeld surface anal. and their 2D fingerprint plots. The most significant contributions in both the compounds are due to H···H interactions which also account for their stability.

A simple, efficient and rapid practical procedure for the preparation of various thiazole derivatives was developed. The present protocol offered the solid phase reaction of various α-bromo ketones with a wide variety of thioamides/thioureas in the presence of Na2CO3 at room temperature (within 3-5 min). More specifically, it also provided an excellent synthetic route to 2-(pyrazol-1-yl)thiazole derivatives that can not be prepared easily using solvent or acidic conditions mediated Hantzsch thiazole reaction due to the exclusive formation of thiocyanatoketones. Increase in amount of solid catalyst led to the formation of 4-hydroxythiazole, a cyclic intermediate which is formed in Hantzsch thiazole reaction. In conclusion, reaction could be stopped either at intermediate state or was directed towards thiazole formation by changing the amount of base.
