A new series of benzotriazole moiety bearing substituted imidazol-2-thiones I [R = H, 4-Me, 2-F, etc.] at N1 was designed, synthesized and evaluated for in vitro anticancer activity against the different cancer cell lines MCF-7(breast cancer), HL-60 (Human promyelocytic leukemia), and HCT-116 (colon cancer). Most of the benzotriazole analogs exhibited promising antiproliferative activity against tested cancer cell lines. Among all the synthesized compounds, I [R = 2,4-di-Cl] showed potent activity against the cancer cell lines such as MCF-7, HL-60 and HCT-116 with IC50 3.57, 0.40 and 2.63μM, resp. Compound I [R = 2,4-di-Cl] was taken up for elaborate biol. studies and the HL-60 cells in the cell cycle were arrested in G2/M phase. Compound I [R = 2,4-di-Cl] showed remarkable inhibition of tubulin polymerization with the colchicine binding site of tubulin. In addition, compound I [R = 2,4-di-Cl] promoted apoptosis by regulating the expression of pro-apoptotic protein BAX and anti-apoptotic proteins Bcl-2. These results provide guidance for further rational development of potent tubulin polymerization inhibitors for the treatment of cancer.

The oxidation product in each reaction is found to be the corresponding phenylurea. The rate of reaction is dependent on [CTAP], [phenylthiourea], [surfactant] and reaction temperature A Michaelis-Menten type kinetics is observed with respect to the substrate. A reaction mechanism involving the formation of a cyclic intermediate in the slow step is proposed for the oxidation reaction.

Mechanochem. solid-state synthesis of 2-aminothiazoles, 2-hydrazinylthiazoles, quinoxalines and benzoylbenzofurans via one-pot sequential α-chlorination of ketones followed by base-mediated condensations with thiourea/thiosemicarbazides, o-phenylenediamine and salicylaldehyde resp. under ball-milling conditions was described. Chlorination of ketones proceeded with atom-economical trichloroisocyanuric acid reagent in the presence of p-TSA under ball-milling conditions to give α-chloroketones which was then further used without isolation. The viability of one-pot sequential acid- and base-mediated reactions in the solid state under ball-milling conditions was thus demonstrated.

Antiviral drug resistance in influenza infections has been a major threat to public health. To develop a broad-spectrum inhibitor of influenza to combat the problem of drug resistance, we previously identified the highly conserved E339…R416 salt bridge of the nucleoprotein trimer as a target and compound 1 as an inhibitor disrupting the salt bridge with an EC50 = 2.7μM against influenza A (A/WSN/1933). We have further modified this compound via a structure-based approach and performed antiviral activity screening to identify compounds 29 and 30 with EC50 values of 110 and 120 nM, resp., and without measurable host cell cytotoxicity. Compared to the clin. used neuraminidase inhibitors, these two compounds showed better activity profiles against drug-resistant influenza A strains, as well as influenza B, and improved survival of influenza-infected mice.

Especially, the analog II (R1 = F; R2 = H; R3 = NH2) exhibited approx. potency with AZD5438 toward four cells including HeLa, HCT116, PC-3, and MDA-MB-231 with IC50 values of 0.45, 0.70, 0.92, 1.80 μM, resp. More interestingly, the most highly active compound II (R1 = F; R2 = H; R3 = NH2) in this study also possessed promising CDK2/cyclin A2 inhibitory activities with IC50 values of 15.4 nM, which was almost 3-fold potent than pos. control AZD5438, and mol. docking studies revealed that the analog bound efficiently with the CDK2 binding site. Further studies indicated that compound II (R1 = F; R2 = H; R3 = NH2) could induce cell cycle arrest and apoptosis in a concentration-dependent manner. These observations suggest that pyrimidine-benzothiazole hybrids represent a new class of CDK2 inhibitors and well worth further investigation aiming to generate potential anticancer agents.
