Using a Diels-Alder reaction, an anthracene unit with four functionalized alkyl substituents (“legs”) was coupled to maleimide-functionalized mol. switches or motors under ambient conditions. Terminal functional groups at the “legs” include thioacetates and azides, making these switches and motors ideal candidates for attachment to metallic or alkyne-functionalized surfaces. UV/vis absorption spectroscopy shows that the mol. switches and motors retain their ability to undergo reversible photoinduced and/or thermally induced structural changes after attachment to the tetrapodal anthracene.

Photochromic cholinesterase inhibitors were obtained from cis-1,2-α-dithienylethene-based compounds by incorporating one or two aminopolymethylene tacrine groups. All target compounds are potent acetyl- (AChE) and butyrylcholinesterase (BChE) inhibitors in the nanomolar concentration range. Compound I bearing an octylene linker exhibited interactions with both the catalytic active site (CAS) and the peripheral anionic site (PAS) of AChE. Yet upon irradiation with light, the mechanism of interaction varied from one photochromic form to another, which was investigated by kinetic studies and proved “photoswitchable”. The AChE-induced β-amyloid (Aβ) aggregation assay gave further exptl. support to this finding: Aβ1-40 aggregation catalyzed by the PAS of AChE might be inhibited by compound I in a concentration-dependent manner and seems to occur only with one photochromic form. Computational docking studies provided potential binding modes of the compound Docking studies and mol. dynamics (MD) simulations for the ring-open and -closed form indicate a difference in binding. Although both forms can interact with the PAS, more stable interactions are observed for the ring-open form based upon stabilization of a water mol. network within the enzyme, whereas the ring-closed form lacks the required conformational flexibility for an analogous binding mode. The photoswitchable inhibitor identified might serve as valuable mol. tool to investigate the different biol. properties of AChE as well as its role in pathogenesis of AD in in vitro assays.

The bisthienylethene-rhodamine 6G dyad exhibit excellent phtochromism with reversibly color and fluorescence changes alternating irradiation with UV and visible light. Upon addition of Hg2+, its color changes from colorless to red and its fluorescence is remarkably enhanced. Whereas other ions including K+, Na+, Ca2+, Mg2+, Fe2+, Co2+, Ni2+, Cu2+, Zn2+, Mn2+, Pb2+, Ni2+, Fe3+, Al3+, Cr3+ and so on induce basically no spectral changes, which constitute a highly selective and sensitive photoswitchable fluorescent probe toward Hg2+. Furthermore, by means of laser confocal scanning microscopy experiments, it is demonstrated that this probe can be applied for live cell imaging and monitoring Hg2+ in living lung cancer cells with satisfying results, which shows its value of potential application in environmental and biol. systems.

Current development of light-responsive materials and technologies imposes an urgent demand on visible-light photoswitching on account of its mild excitation with high penetration ability and low photo-toxicity. However, complicated mol. design and laborious synthesis are often required for visible-light photoswitch, especially for diarylethenes. Worse still, a dilemma is encountered as the visible-light excitation of the diarylethene is often achieved at the expense of photochromic performances. To tackle these setbacks, we introduce a building-block design strategy to achieve all-visible-light photochromism with the triplet-sensitization mechanism. The simply designed diarylethene system is constructed by employing a sensitizer building-block with narrow singlet-triplet energy gap (ΔEST) to a diarylethene building-block. A significant improvement on the photochromic efficiency is obtained as well as an enhanced photo-fatigue resistance over those under UV irradiation The balance between the visible-light excitation and decent photochromism is thus realized, promoting a guiding principle for the visible-light photochromism.

Photopharmacol. involving azobenzene has offered a viable alternative for combating bacterial resistance. However, the degradation and potential toxicity of azobenzene limit its further study in vivo. Therefore, searching for an appropriate photoswitch for further clin. application is highly desirable. Herein a series of dithienylethene-functionalized switchable antibacterial agents have been designed and prepared by the introduction of the dithienylethene scaffold into fluoroquinolones. And it was found that these switchable antibacterial agents displayed good photochromism and fluorescence switching behaviors upon irradiation with UV/Vis light in DMSO. Surprisingly, methoxy-substituted dithienylethenes 3a and 3b exhibited fluorescence turn-on behavior. Furthermore, it was found that all of the open-isomers showed partial antibacterial activity on E. coli and S. aureus compared with the native drugs. Apart from 2a and 2b, the other switchable antibacterial agents showed a large difference in antibacterial activity on Gram-neg. E. coli between the open and closed forms, in which the antimicrobial activity of the ring-closed isomers for 1b and 3b was 16 times that of the corresponding ring-open isomers. DFT calculations showed that the ring-closed isomers of 1b and 3b presented a rigid “S-type” conformation, which may be conducive to forming more stable complexes with the DNA gyrase of E. coli.

Here, a novel D-DTE-A type dithienylethene NDB, which was modified by difluoroboron β-diketonate (BF2bdk) and dimethylphenylamine (DMPA) moieties, has been developed. It exhibited solvent-dependent photophys. properties before irradiation, and the visible light-triggered NIR photochromic behavior with solvent-dependent features was only observed in Toluene. Moreover, it displayed intense fluorescent turn-on acid-sensing properties in less polar solvent [Toluene, Dichloromethane (DCM) and Et acetate (EA)] through the blockage of photoinduced electron transfer (PET) effect. Three rationally designed axially chiral diarylethene switches were synthesized and their application as chiral dopants for phototunable cholesteric liquid crystal devices was investigated. Design of these mols. was based on the combination of photochromic dithienylcyclopentene core with bridged binaphthyl units as chiral precursors. Aromatic groups were introduced to the mols. at 6,6′-positions of binaphthyls through a Suzuki-Miyaura coupling reaction. Their helical twisting powers (HTPs) are significantly higher than those of the known chiral diarylethenes reported as chiral dopants so far. Photocyclization of these mols. upon light irradiation brought out dramatic variation in HTPs between different states. The primary colors, red, green, and blue, were obtained in reflection on light irradiation and with thermal stability. Moreover, a multi-switchable photodisplay was demonstrated using one of these chiral mol. switches.
