Adenylate cyclases (ACs), play a critical role in the conversion of ATP (ATP) into the second messenger cyclic adenosine monophosphate (cAMP). Studies have indicated that adenylyl cyclase type 2 (AC2) is potential drug target for many diseases, however, up to now, there is no AC2-selective agonist reported. In this research, docking-based virtual screening with the combination of cell-based biol. assays have been performed for discovering novel potent and selective AC2 agonists. Virtual screening disclosed a novel hit compound 8 as an AC2 agonist with EC50 value of 8.10μM on recombinant human hAC2 + HEK293 cells. The SAR (structure activity relationship) based on the derivatives of compound 8 was further explored on recombinant AC2 cells and compound 73 was found to be the most active agonist with the EC50 of 90 nM, which is 160-fold more potent than the reported agonist Forskolin and could selectively activate AC2 to inhibit the expression of Interleukin-6. The discovery of a new class of AC2-selective agonists would provide a novel chem. probe to study the physiol. function of AC2.

A series of isomeric poly(thioether imide)s (PTIs) containing thioether linkages were prepared by aromatic nucleophilic substitution reaction of isomeric N-(4-mercaptophenyl)-3-chlorophthalimide and/or N-(4-mercaptophenyl)-4-chlorophthalimide. The inherent viscosities of these polymers were in the range of 0.38-0.42 dL/g. The glass transition temperature increased with increasing the content of 3-substituted phthalimide unit in the polymer backbone. The 5 % weight loss temperatures of isomeric PTIs (a-e) were 506-531° in nitrogen and 509-536° in air, resp. The isomeric PTIs (b-d) films exhibited good mech. properties with tensile strength of about 90 MPa, elongation at break of more than 6.0 %, and tensile moduli of more than 2.2 GPa. The storage moduli of PTIs (b-d) almost completely maintained at about 2.0 GPa before reaching the corresponding glass transition temperatures It is noted that the min. melt viscosity of these polymers decreased with increasing the content of unsym. 3- and 4-substituted phthalimide unit.

A novel composite of polythioetherimide (PTEI)/bismaleimide (BMI)/2,2′-diallylbisphenol A (BA) was prepared Mech. measurements, dynamic thermomech. anal., and microanal. were conducted to assess the toughness and morphol. of the composite. Results demonstrate that the addition of PTEI has significant effects on the mech. properties of the BMI/BA system. Compared with the BMI/BA resin, the PTEI/BMI/BA system significantly improves the impact strength and bending strength, given an appropriate amount of PTEI. Scanning electron microscope (SEM) images and energy dispersive spectroscopy results show that PTEI is distributed uniformly in the PTEI/BMI/BA system and that the interfacial compatibility between the PTEI and BMI/BA phase is excellent. Thermogravimetric and dielec. analyses were also conducted to infer the thermal and dielec. properties of the composite. With a suitable addition of PTEI, the BMI/BA resin still exhibits good thermal resistance and dielec. constant and loss values of the blends still retain good stability in a frequency band from 10 MHz to 60 MHz. All these changes in properties are closely correlated with the addition of PTEI.
