A stereoselective gold(I)-catalyzed vinylcyclopropanation of alkenes has been developed. A gold-coordinated cationic vinyl carbene species, readily generated via a rearrangement of the ethylenedithioacetal of propargyl aldehyde, reacts with a wide range of alkenes to afford thio-substituted vinylcyclopropanes. The gold-catalyzed vinyl cyclopropanation proceeds under mild conditions at room temperature and is generally selective for the formation of cis-substituted cyclopropanes. The reaction allows the formal introduction of a “naked” vinyl carbene, by subsequent chemoselective hydrodesulfuration of the ethylenedithio-bridge. The synthetic utility of the new method is demonstrated by a short, racemic formal synthesis of the alkaloid cephalotaxin, hinging on a key vinyl cyclopropane-cyclopentene rearrangement.

Identification of chems. that affect hormone-regulated systems will help to predict endocrine disruption. In our previous study, a 46 gene biomarker was found to be an accurate predictor of estrogen receptor (ER) α modulation in chem. treated MCF-7 cells. Here, potential ERα modulators were identified using the biomarker by screening a microarray compendium consisting of ∼1600 gene expression comparisons representing exposure to ∼1200 chems. A total of ∼170 chems. were identified as potential ERα modulators. In the Connectivity Map 2.0 collection, 75 and 39 chems. were predicted to activate or suppress ERα, and they included 12 and 6 known ERα agonists and antagonists/selective ERα modulators, resp. Nineteen and 8 of the total number were also identified as active in an ERα trans-activation assay carried out in a MCF-7-derived cell line used to screen the Tox21 10K chem. library in agonist or antagonist modes, resp. Chems. predicted to modulate ERα in MCF-7 cells were examined further using global and targeted gene expression in wild-type and ERα-null cells, trans-activation assays, and cell-free ERα coregulator interaction assays. Environmental chems. classified as weak and very weak agonists were confirmed to activate ERα including apigenin, kaempferol, and oxybenzone. Novel activators included digoxin, nabumetone, ivermectin, and six progestins. Novel suppressors included emetine, mifepristone, niclosamide, and proscillaridin. Our strategy will be useful to identify environmentally relevant ERα modulators in future high-throughput transcriptomic screens.

This paper evaluates the 5-aryl-2-pyridyloxazole backbone to engineer donor-acceptor fluorescent pH probes after one- or two-photon absorption. Parent fluorophores, as well as derivatives that can be used to label biomols., can be easily obtained in good yields. These mols. exhibit a large one-photon absorption in the near-UV range, and a strong fluorescence emission that covers the whole visible domain. The 5-aryl-2-pyridyloxazole derivatives also possess significant cross sections for two-photon absorption. Upon pyridine protonation, large shifts were observed in the absorption spectra after one- and two-photon excitation, as well as in the emission spectra. This feature was used to measure the pKa of the investigated compounds that range between 2 and 8. In most of the investigated derivatives, the pKa increased upon light excitation and protonation exchanges took place during the lifetime of the excited state, as shown by phase-modulation fluorometry anal. Several 5-aryl-2-pyridyloxazole derivatives are suggested as efficient probes to reliably measure the pH of aqueous solutions by means of ratiometric methods that are dependent on fluorescence emission.

The paucity of specific pharmacol. agents has been a major impediment for delineating the roles of gap junction (GJ) channels formed by connexin proteins in physiol. and pathophysiol. Here, we used the selective optimization of side activities (SOSA) approach, which has led to the design of high affinity inhibitors of other ion channels, to identify a specific inhibitor for channels formed by Cx50, a connexin subtype that is primarily expressed in the lens. We initially screened a library of common ion channel modulating pharmacophores for their inhibitory effects on Cx50 GJ channels and identified four new classes of compounds The triarlymethane (TRAM) clotrimazole was the most potent Cx50 inhibitor and we therefore used it as a template to explore the structure activity relationship (SAR) of the TRAMs for Cx50 inhibition. We describe the design of T122 (N-[(2-methoxyphenyl)diphenylmethyl]-1,3-thiazol-2-amine) and T136 (N-[(2-iodophenyl)diphenylmethyl]-1,3-thiazol-2-amine), which inhibit Cx50 with IC50s of 1.2 and 2.4 μM. Both compounds exhibit at least 10-fold selectivity over other connexins as well as major neuronal and cardiac voltage-gated K+ and Na+ channels. The SAR studies also indicated that the TRAM pharmacophore required for connexin inhibition is significantly different from the pharmacophore required for blocking the calcium-activated KCa3.1 channel. Both T122 and T136 selectively inhibited Cx50 GJ channels in lens epithelial cells, suggesting that they could be used to further explore the role of Cx50 in the lens. In addition, our results indicate that a similar approach may be used to find specific inhibitors of other connexin subtypes.

The influence of eight different quaternary ammonium and phosphonium salts on the vulcanization of natural rubber was investigated. The central atom (P or N) of the cation was of minor importance, but the nature of the organic chains greatly affected the rheometrical parameters of the process and the mech. properties of the vulcanizates. Studies of rubber compositions containing binary mixtures of a quaternary salt and the com. accelerators ZDEC, TBBS, TMTD and MBTS showed a synergistic effect, mainly with TBBS and MBTS. With such binary systems, it is possible to reduce the amount of accelerator and improve the vulcanization parameters and the quality of the vulcanized rubber. The anionic complex bis(N-phenylsulfonyldithiocarbimato)zincate(II) (Z), one amine-free exptl. accelerator, was prepared and isolated as tetrabutylammonium (B2Z) and benzyltriphenylphosphonium (H2Z) salts. The H2Z complex salt is a new substance and behaved as an ultra-accelerator, being faster than ZDEC and TMTD. B2Z was a medium rate accelerator, being faster than TBBS and similar to MBTS. In addition, the tensile and tear strength of the specimens vulcanized with the zinc-dithiocarbimate salts were superior when compared to compositions containing the com. accelerators. The differences in the behavior of B2Z and H2Z showed that the zinc-dithiocarbimates are versatile accelerators for natural rubber as the process and the quality of the vulcanizate can be modulated by the choice of the cation.

According to IR spectroscopy, under the influence of the biol. medium 199 there are processes of biodegradation of polymer materials which are accompanied by a decrease in the intensity of the absorption band νC=O of vinylpyrrolidone ring. It is established that the introduction of tiamulin fumarate into polyurethaneurea facilitates their biodegradation, as evidenced by a decrease of the tensile strength (1.28-2.38 times) and a relative elongation at break (1.36-1.82 times) after incubation in the biol. medium 199 by physico-mech. tests. The tissue culture method has established the biocompatibility of film materials with tiamulin fumarate. According to the results of microbiol. studies, bactericidal properties were observed in relation to the bacteria Staphylococcus aureus and Escherichia coli. It allows using them for production of drainage in abdominal surgery.
