Finally, this compound also demonstrated favorable in vitro ADME and pharmacokinetic properties (e.g. Cmax = 5.7μM, Tmax = 15 min, t1/2 = 3.4 h and Cl = 45 mL/min/kg following single 10 mg/kg i.p. administration) further supporting the use of pyrido[2,3-d]pyrimidin-7-ones as a new structure class of RIPK2 kinase and NOD cell signaling inhibitors.

The solute -solvent interaction between ionic liquids (ILs) and amino acids (AA) in aqueous media plays a significant role for the optimization of a number of important biotechnol. processes. L-Valine and L-Proline (two solute mols.) interact with an ionic liquid (Benzyltributylammonium chloride) in aqueous medium. Based on the different parameters such as apparent molar volume, viscosity B-coefficient, molar refraction, molar conductance at different temperatures and different concentrations from d., viscosity, refractive index, conductance measurements have been used to explain the mol. level interactions which was supported by NMR and UV-Vis studies. Using Masson equation, the exptl. slopes and the limiting apparent molar volumes are obtained which explain the solute-solute and solute-solvent interactions. Hepler’s technique and dB/dT values have been used to examine the structure-making and structure-breaking nature of the solutes in the solvents. Viscosity parameters, A and B obtained from Jones-Doles equation explained the solute-solute and solute-solvent interactions in the solution Lorentz-Lorenz equation has used to calculate the molar refraction. The specific conductance also explained the interaction properties. Further the findings have been supported by NMR study of the solutions and also considerable amount of theor. anal. has been done which was in good agreement with the exptl. result. The behavior of many other bio-mols. can be explained by considering amino acids as model and the mechanism has been extended to elucidate the behavior of other (biol.) systems. In our findings we were emphasized on the nature of solute-solvent interactions and the presence of structural effect on the solvent in solution to analyze the mol.-level interactions prevalent in the systems.

Alzheimer’s disease (AD) is the most prevalent disease of old age leading to dementia. Complex AD pathogenesis involves multiple factors viz. amyloid plaque formation, neurofibrillary tangles and inflammation. Herein we report of a new series of quinoxaline-bisthiazoles as multitarget-directed ligands (MTDLs) targeting BACE-1 and inflammation concurrently. Virtual screening of a library of novel quinoxaline-bisthiazoles was performed by docking studies. The most active mols. from the docking library were taken up for synthesis and characterized by spectral data. Compounds 8a-8n showed BACE-1 inhibition in micro molar range. One of the compounds, 8n showed BACE-1 inhibition at IC50 of 3 ± 0.07 μM. Rat paw edema inhibition in acute and chronic models of inflammation were obtained at 69 ± 0.45% and 55 ± 0.7%, resp. Compound 8n also showed noteworthy results in AlCl3 induced AD model. The treated rats exhibited excellent antiamnesic, antiamyloid, antioxidant, and neuroprotective properties. Behavioral parameters suggested improved cognitive functions which further validates the testimony of present study. Moreover, compound 8n was found to have inherent gastrointestinal safety. This new string of quinoxaline-bisthiazoles were identified as effective lead for the generation of potent MTDLs and compound 8n was found to showcase qualities to tackle AD pathogenesis.

The surface structures of the bonded phases are determined by 29Si and 13C cross-polarization and magic angle spinning NMR. Furthermore, the retention behavior of the phases is investigated with polar and nonpolar solutes, and data are correlated in terms of the bonding procedure and the functionality of the silanizing agent. Mass loading experiments for a single solute on selected packings give a maximum load of about 140.0 and 70.0 μg/g of packing for large and small particles, resp., irresp. of the silanizing agent.

In comparison with the organic solvent process, the fluidized bed technique produces a more reproducible packing regardless of the functionality of the silanizing agent.

The effect of temperature and mobile phase composition (methanol-water) on the retention behavior of an oligomeric series of n-octylsilyl bonded phases in reversed-phase liquid chromatog. was investigated. The ΔH° values of the aromatic hydrocarbons and n-alkyl benzoates are higher than those of the polar compounds due to their higher level of interaction with the stationary phase. A linear plot of ΔH° vs. ΔS° suggest that the retention process, which is essentially controlled by non-specific (dispersive) interactions between the solutes and the bonded ligands, is identical for all cases evaluated. The existence of similar retention mechanisms is confirmed by the constant value of the enthalpy-entropy compensation temperature of the columns for a given class of compounds As expected, decreasing the methanol content (% volume/volume) of the mobile phase results in increased eluate retention times. The methylene and Ph selectivities are found to be independent of the carbon content of the stationary phases and varied only with the eluent composition In addition to their high stability under aggressive mobile phase conditions as previously reported, the results of this study generally showed that the solute retention process on oligomeric phases are similar to those exhibited by the conventional reversed phases.
