Profiling the occurrence of biogenic amines in different types of tuna samples using an improved analytical approach

Food deterioration caused by microbial agents often involve the formation of biogenic amines (BAs), which can have harmful effects on human health. In this study a set of BAs – tryptamine, cadaverine, putrescine, spermine, histamine, tyramine, and spermidine, were simultaneously analysed to monitor their occurrence in different types of tuna samples. An improved extraction approach involving ultrasound-assisted microextraction (USA mu ET) followed by derivatization with dansyl chloride (DnsCl) and analysis by ultrahigh performance liquid chromatography (UHPLC) with fluorescence detection was validated for BAs quantification. The performance of the USA mu ET/UHPLC-FLD was assessed by studying the limits of detection (LOD) and quantification (LOQ), linear dynamic range (LDR), precision (intra and inter-day) and matrix effect (ME). Good linearity (r(2) > 0.98), LODs (from 0.98 to 8.57 mg kg(-1)) and LOQs (3.20-25.6 mg kg(-1)) were achieved for all BAs analysed. Recoveries ranged from 76% to 106%, with relative standard deviations (RSD) lower than 5.0%. ME was determined from 7.52 to 50% and the intra and inter-day precisions ranged from 4.7 to 11.6% and 5.5-14.2%, respectively. BAs levels varied significantly from 4.09 +/- 0.8 mg kg(-1) of putrescine in olive oil canned samples to 577.9 +/- 5.9 mg kg(-1) of cadaverine in natural canned samples. Tryptamine and tyramine were not detected in any of the samples analysed, while spermine and spermidine were found in 85.7% of the assayed tuna samples. Cadaverine was the most dominant BA with concentrations ranging from 54.3 +/- 2.5 mg kg(-1), in olive oil canned tuna, to 577.9 +/- 5.9 mg kg(-1), in natural canned samples, whereas putrescine had the lowest concentration (average 6.9 +/- 2.5 mg kg(-1)). The validated methodology revealed important improvements in terms of simplification of the experimental layout, expressed in the low sample and reagent amounts, in addition to less time-consuming and labour-intensive requirements that did not compromise the analytical performance.

Our mice’s findings demonstrate that intracerebroventricular injection of EGTA enhances the sevoflurane-induced loss of righting reflex ED50 while injecting calcium-40 chloride or calcium-43 chloride causes an opposite effect. Further, the identical effects of calcium-40 and calcium-43 indicate an absence of calcium isotope dependence. Here, our findings disprove conventional proposals that calcium ion concentration correlates with consciousness.

Effect of ultra-fine fly ash on concrete performance and durability

In the present study ultra-fine fly ash as a novel by-product obtained by a dry and closed separation process was investigated as cement replacement in concrete. The impact of ultra-fine fly ash on material properties was investigated following an upscaling as an approach considering paste, mortar and concrete properties. Two types of cement, Portland cement (CEMI) and slag cement (CEMIII), two types of ultra-fly ashes, one with particle size d(90) < 9.3 mu m (FA1) and second with d(90) < 4.6 mu m (FA2) were used. At paste- and mortar- level, cement was replaced at 0%, 15%, 25%, 35% and 50% with FA1 and FA2. At concrete- level, cement was replaced at 0%, 15% and 25% with different ratios of FA1 and FA2. The results at paste- and mortar- level showed that an increased fineness of the fly ash (FA2) contributes to better workability of the mix. For CEMI, the compressive strength of concrete with FA2 at 25% cement replacement was already equal to the reference 0% replacement concrete at the age of 28 days. For CEMIII, the compressive strength of concrete mix with FA1 with 15% and FA2 with 25% cement replacements reached the reference concrete value at the age of 91 days. Regarding the durability, replacing cement with ultrafine fly ash (FA2) had a positive influence on the resistivity, chloride migration coefficient and alkali-silica reaction (ASR), and a negative influence on the carbonation resistance. An experimental procedure is presented that was developed by fifth-year chemical engineering and industrial chemistry undergraduates at the Federal University of Rio de Janeiro doing the discipline on Experimental Organic Technology. The aim of this study was to apply the solvatochromic effect of the dye Nile Blue chloride to the characterization of biodiesel/diesel blends with different biodiesel content, using an alternative image processing analysis method involving low-cost, simple, and rapid assays. The effect of the solvatochromic dye Nile Blue chloride was monitored using the software ImageJ, which is being used currently, yielding rapid and effective responses. As an outcome, the students proposed a procedure that had potential application to colorimetric determination of biodiesel content in diesel oil. This gave students the opportunity to put the knowledge acquired in the course into practice, by dealing with issues concerning the fuels industry, making the learning process more dynamic, engaging, and effective.

Oligomerization and Polymerization of Isoprene Catalyzed by Alkylaluminium with Different Structures

Alkylaluminium (AlR3), as co-catalyst component in Ziegler-Natta catalytic system, plays important roles in the alkylation, forming and changing the structure and concentration of active centers through the reduction and reversible adsorption-desorption reactions with the metal compound of the catalyst, acting as chain transfer agent, etc. However, the alkylaluminium itself do have the catalytic effect on the conjugated diene monomers. In this article, alkylaluminium with different structures such as triethylaluminium (AlEt3), triisobutylaluminium (Al(i-Bu)(3)), diisobutylaluminium hydride (AlH(i-Bu)(2)), diethylaluminium chloride (AlEt2Cl), ethylaluminium dichloride (AlEtCl2) were used to catalyze isoprene oligomerization and polymerization. The effects of the structure and concentration of alkylaluminiums (n(Al)/n(M)=7 x 10(-5), 35 x 10(-5), 350 x 10(-5), 1050 x 10(-5)) on the catalytic behaviors of isoprene were studied. The microstructure (trans-1,4 and cis-1,4), molecular weight and molecular weight distribution of the products were characterized by H-1 nuclear magnetic resonance spectroscopy (H-1 NMR), gel permeation chromatography (GPC) and gas chromatography-mass spectrometry (GC-MS). It was found that alkylaluminium could initiate oligomerization and cationic polymerization of isoprene under the minor incorporation of H2O, which were affected greatly by the structure and concentration of alkylaluminium. Using AIEtCl(2) led to the highest catalytic activity and produced products containing more linear polymers with mixed cis-1,4/trans-1,4 structures when n(Al)/n(M)=1050 x 10(-5). The Al(i-Bu)(3) and AlH(i-Bu)(2) didn’t have basically cation initiation ability, which led to isoprene oligomerization. The alkylaluminium with n(Al)/n(M)<= 350 x10(-5) had negligible influence on the isoprene polymerization and oligomerization. And lower or higher alkylaluminium concentration were not beneficial to obtain polyisoprene with high molecular weight. The catalytic mechanism of alkylaluminium on isoprene was discussed, which provided a further understanding on the catalytic behavior of alkylaluminium components in Ziegler-Natta catalyst and the effect of alkylaluminium on polymers. Experimental mapping of a pH gradient from a positively charged micellar interface to bulk solution

The nanoscale pH gradients from a charged membrane interface to the bulk solution, determine fundamental properties of chemical and biological systems. We estimated the proton distribution on micelle surfaces experimentally measuring the apparent pK(a) of a weak acid linked at varying distances to a micelle anchoring moiety and relating these values to the electrical potential at the micellar surface. The pH probes consisted of benzoic acid connected to chains with variable number (1-5) oxyethylene (-OCH2CH2-) units. The methylene of the terminal oxyethylene was connected to a quaternary nitrogen containing a long hydrocarbon chain. We calculated the apparent pK(a) (pK(ap)) of the benzoic acid derivatives with surfactant micelles by UV spectroscopy. The difference between the pK(a) in solution and the pK(ap) in micelles (Delta pK) of all probes was very similar with negatively charged sodium dodecyl sulphate (SDS) or zwitterionic N-hexadecyl-N,N-dimethyl-3-ammonio-1-pmpanesulfonate (HPS) micelles, indicating that the dissociation of the substituted benzoates occurred at the micellar interface in both SDS and HPS. In contrast, the Delta pK of the probes decreased as a function of the benzoate’s distance to the micelle interface in hexadecyltrimethylammonium chloride (CTAC) micelles, The variation of Delta pK with distance, from the CTAC micellar interface to bulk solution, was quantitatively correlated to H+ distribution from the interface. The experimental results coincided with independently calculated Poisson Boltzmann H+ distribution. Pt nanoparticle decorated carbon nanotubes nanocomposite based sensing platform for the monitoring of cell-secreted dopamine

Development of reliable sensors for real-time quantification of dopamine is highly essential to understand various biological processes and still remains challenge. Here, we proposed a novel and ultrasensitive electrochemical sensing platform for the monitoring of cell-secreted dopamine based on Pt nanoparticles (PtNPs) decorated multi-wall carbon nanotubes (MWCNTs) nanocomposite, which is synthesized by in-situ reduction of platinum on poly (diallyldimethylammonium chloride) (PDDA) functionalized MWCNTs. The unique structure of the hybrid nanocomposite and the interconnectivity between PtNPs and carbon nanotubes endow the nanocomposite with the enhancing conductivity and electrochemical activity. The Pt-MWCNTs hybrid modified screen-printed electrode exhibited excellent electrocatalytic ability for the detection of dopamine with a wide linear range (5 nM to 1 mu M) and a low detection limit (2 nM). The developed sensor enables the real-time recording of dopamine released from PC12 cells induced by the stimulation of K+ saline. Also, this approach can be used to quantitatively evaluate the effects of antipsychotic drug (aripiprazole) on the dopamine release from cells treated with high K+. Results indicated that the sensor could provide an effective tool for the monitoring of extracellular neurotransmitters related to the exogenous agent and drug on the nervous system.

Adsorptivity of cationic cellulose nanocrystals for phosphate and its application in hyperphosphatemia therapy

Nanocellulose has gained much attention because of its excellent properties. Cationic cellulose nanocrystals (cCNC) shows good adsorptivity toward negative ions and molecules. Phosphate binders are most used to treat hyperphosphatemia and it is significant to develop its alternatives with high specific and low cost in the clinic. Herein, we prepared cCNC and characterized it by FTIR, TEM, dynamic light scattering, and viscosity method. We simulated the binding process of cationic cellulose for phosphate and used it as phosphate binder for hyperphosphatemia therapy to study the phosphate binding effect and evaluate the oral toxicity. Cationic cellulose improved the conditions of mice models and efficiently decreased the level of phosphate in the serum. cCNC had a better binding effect than cationic micmcrystalline cellulose both in vitro and in vivo. cCNC could be used as alternatives to phosphate binder for therapy of chronic renal failure and hyperphosphatemia.

Herb pair of Ephedrae Herba-Armeniacae Semen Amarum alleviates airway injury in asthmatic rats

Ethnopharmacological relevance: Ephedrae Herba (EH, Ephedra sinica Stapf.) and Armeniacae Semen Amarum (ASA, Prunus armeniaca L. var. ansu Maxim.) have been used to treat asthma, cold, fever, and cough in China for thousands of years. Aim of the study: In this study, we aimed to investigate the optimal ratio of EH and ASA compatibility (EAC) to reduce airway injury in asthmatic rats and its possible mechanism. Methods: Rats were sensitized with a mixture of acetylcholine chloride and histamine bisphosphate 1 h before sensitization by intragastric administration of EAC or dexamethasone or saline for 7 days. Subsequently, the ultrastructure of rat airway epithelial tissue changes, apoptosis of the airway epithelial cells, and the expression of mRNA and protein of EGRF and Bcl-2 were detected. Results: Transmission electron microscope: EAC (groups C and E) had the most prominent effect on repairing airway epithelial cells’ ultrastructural changes in asthmatic rats. TUNEL: dexamethasone and EAC (groups B, C, E and F) inhibited the apoptosis of airway epithelial cells in asthmatic rats (P < 0.05). In situ hybridization: EAC (group E) inhibited the overexpression of EGFR and Bcl-2 mRNA (P < 0.05).Western Blotting: EAC (groups A, B, C, E and F) inhibited the upregulation of airway epithelial EGFR and Bcl-2 protein expression (P < 0.01). Conclusions: Our findings indicate that EAC can inhibit abnormal changes in airway epithelial structure and apoptosis of airway epithelial cells, thereby alleviating airway injury. In this study, the best combination of EH and ASA to alleviate airway epithelial injury in asthmatic rats was group E (EH: ASA = 8: 4.5). Do you like my blog? Insights into Primary Ion Exchange between Ion-Selective Membranes and Solution. From Altering Natural Isotope Ratios to Isotope Dilution Inductively Coupled Plasma Mass Spectrometry Studies

Although ion-selective electrodes have been routinely used for decades now, there are still gaps in experimental evidence regarding how these sensors operate. This especially applies to the exchange of primary ions occurring for systems already containing analyte ions from the pretreatment step. Herein, for the first time, we present an insight into this process looking at the effect of altered ratios of naturally occurring analyte isotopes and achieving isotopic equilibrium. Benefiting from the same chemical properties of all isotopes of analyte ions and spatial resolution offered by laser ablation and inductively coupled plasma mass spectrometry, obtaining insights into primary ion diffusion in the preconditioned membrane is possible. For systems that have reached isotopic equilibrium in the membrane through ion exchange and between the membrane phase and the sample, quantification of primary ions in the membrane is possible using an isotope dilution approach for a heterogeneous system (membrane-liquid sample). Experimental results obtained for silver-selective membrane show that the primary ion diffusion coefficient in the preconditioned membrane is close to (6 +/- 1) x 10(-)(9) cm(2)/s, being somewhat lower compared to the previously reported values for other cations. Diffusion of ions in the membrane is the rate limiting step in achieving isotopic exchange equilibrium between the ion-selective membrane phase and sample solution. On the contrary to previous reports, quantification of silver present in the membrane clearly shows that contact of the membrane with silver nitrate solution of concentration 10(-3) M leads to pronounced accumulation of silver ions in the membrane, reaching almost 150% of ion exchanger amount. The magnitude of this effect increases for higher concentration of the electrolyte in the solution.
