Abstract: A novel series of 1-(5-(benzylthio)-1,3,4-thiadiazol-2-yl)-3-phenylurea derivatives (5a-l) were designed and synthesized as sorafenib analogs. The in vitro cytotoxicity effects of synthesized compounds were evaluated against four different human cancer cells including MCF-7, HepG2, A549, and HeLa cell lines. The biol. results showed that most of the compounds significantly prevented the proliferation of tested cancer cells. In particular, 2-F, 4-Cl, and 2,6-diF substituted derivatives (5d, 5g, and 5k) showed promising activities, especially against Hela cancer cells (IC50 = 0.37, 0.73 and 0.95 μM, resp.) which were significantly more potent than sorafenib as the reference drug (IC50 = 7.91 μM). Flow cytometry anal. revealed that the prototype compounds (5d, 5g, and 5k) significantly induced apoptotic cell death in HeLa cancer cells and blocked the cell cycle at the sub-G1 phase. Moreover, in silico docking study confirmed the binding of the prototype compound to the active site of VEGFR-2.

A series of 5,8-disubstituted tetrahydroisoquinolines e.g., 2-((5-(4-chlorophenyl)pyridin-2-yl)methyl)-8-(4-methylpiperazin-1-yl)-1,2,3,4-tetrahydroisoquinoline were shown to be effective inhibitors of M. tb in culture and modest inhibitors of M. tb ATP synthase. There was a broad general trend of improved potency with higher lipophilicity. Large substituents (e.g., Bn) at the tetrahydroquinoline 5-position were well-tolerated, while N-methylpiperazine was the preferred 8-substituent. Structure-activity relationships for 7-linked side chains showed that the nature of the 7-linking group was important; -CO- and -COCH2– linkers were less effective than -CH2– or -CONH- ones. This suggests that the positioning of a terminal aromatic ring is important for target binding. Selected compounds showed much faster rates of microsomal clearance than the clin. ATP synthase inhibitor bedaquiline, and modest inhibition of mycobacterial ATP synthase.

Diastereofacial selectivity in an enantiospecific intramol. 1,3-dipolar addition is controlled by adjusting the size of the tether between the dipole and the dipolarophile to give 2,3-disubstituted pyrrolidines enantiomeric with respect to the newly generated stereogenic 2,3-centers depending on the tether size; this leads to stereocontrolled synthesis of the (-)-(I) and (+)-enantiomers of the necine base dihydroxyheliotridane from chiral O-benzylglycidol.

Imanishi, Masashi published the artcileDiscovery of a Novel Series of Biphenyl Benzoic Acid Derivatives as Potent and Selective Human β3-Adrenergic Receptor Agonists with Good Oral Bioavailability. Part I, Category: chlorides-buliding-blocks, the publication is Journal of Medicinal Chemistry (2008), 51(6), 1925-1944, database is CAplus and MEDLINE.

A novel class of biphenyl analogs containing a benzoic acid moiety based on lead compound I have been identified as potent and selective human β3 adrenergic receptor (β3-AR) agonists with good oral bioavailability and long plasma half-life. After further substituent effects were investigated at the terminal Ph ring of lead compound I, it has been discovered that more lipophilic substitution at the R position improved potency and selectivity. As a result of these studies, II and III were identified as the leading candidates with the best balance of potency, selectivity, and pharmacokinetic profiles. In addition, compounds II and III were evaluated to be efficacious for a carbachol-induced increase of intravesical pressure, such as an overactive bladder model in anesthetized dogs. This represents the first demonstrated result dealing with β3-AR agonists.

The “Blue Bottle” experiment was first popularized by Campbell. The reaction consists of a flask about half full of a colorless liquid that, when shaken, turns blue. When left to stand the liquid turns colorless again. The cyclic process – being shaken, turning blue, being left to stand, and turning colorless – can be repeated many times. By making a series of guided observations with this simple but colorful system, the student can determine a rate law and a plausible mechanism for the reaction without ever knowing the contents of the flask. The activation energy of the reaction can also be measured by making observations at various temperatures This popular experiment has been carried out by general chem. students at Valparaiso University since 1963 with great success.

The catalytic enantioselective synthesis of 3-aryl-substituted pyrrolopyrimidines (PPYs), a common motif in drug discovery, is achieved through a kinetic resolution via quaternary ammonium salt-catalyzed nucleophilic aromatic substitution (SNAr). Both enantioenriched products and starting materials can be functionalized with no observed racemization to give enantiodivergent access to diverse chiral analogs of an important class of kinase inhibitor. One of the compounds was found to be a potent and selective inhibitor of breast tumor kinase.

Sanchez, Raquel published the artcileEffect of the silicon chemical form on the emission intensity in inductively coupled plasma atomic emission spectrometry for xylene matrices, COA of Formula: C7H16Cl2Si, the publication is Journal of Analytical Atomic Spectrometry (2009), 24(4), 391-401, database is CAplus.

Silicon is present in petrol products under very different chem. forms. The effect of the silicon chem. form on the sensitivity was studied by inductively coupled plasma at. emission spectrometry (ICP-AES). Solutions containing sixteen different silicon organic compounds with the same silicon concentration were prepared in xylene. The studied compounds were: six siloxanes, nine silanes and a silicate. Their b.ps. ranged from 101° (hexamethyldisiloxane) to 310° (2,3,4-(epoxycyclohexyl)ethyltrimetoxysilane). The ICP-AES signal depended strongly on the silicon compound Thus, for a commonly used sample introduction system consisting of a cross-flow pneumatic nebulizer coupled to a Ryton double pass spray chamber, the signal varied by a factor of up to 17, depending on the compound For most studied cases, some siloxanes yielded higher emission intensities than silanes. Under some circumstances, hexylmethyldichlorosilane and dimethyldodecylchlorosilane also produced higher signals than the other studied silanes. To elucidate the possible mechanisms for these changes, experiments were performed to verify whether the origin could be assigned either to the plasma or to the sample introduction system. Using the Mg II/Mg I ratio, no significant change in the plasma characteristics was actually observed As regards the sample introduction system, (i) the primary aerosol, (ii) the transport and memory effects using continuous and discrete introduction modes, an alternative spray chamber, and a total consumption system (the TISIS system), and (iii) the tertiary aerosol, via an impactor, were studied. Addnl. experiments were carried out with an ultrasonic nebulizer. The Si behavior was compared to other elements, such as Cu, Mg, Mn and Zn. The Si behavior was not related to the pneumatic nebulizer and suggested that a fraction of silicon was transported towards the plasma as a vapor when it was present as hexamethyldisiloxane (i.e., the compound with the lowest b.p. among the species studied). In any case, the variability of the signal induced by the different silicon chem. forms was less significant for the TISIS than for cyclonic and ultrasonic nebulizer. The observed results were specific to silicon as the signal of the other elements did not significantly change with silicon chem. form. As a consequence, internal standardization was not appropriate for mitigating interferences on silicon when petroleum products were analyzed.

It was shown previously that the influence of the silicon chem. form could be minimized in ICP-AES by using high efficiency sample introduction systems. The effects of two of the most different silicon chem. forms were minimized in ICP-AES by using near total sample consumption systems. The two tested devices were a single pass spray chamber equipped with wall heating (heated torch integrated sample introduction system, h-TISIS) and a demountable direct injection high efficiency nebulizer (d-DIHEN). A room temperature TISIS and a cyclonic spray chamber were taken as the reference systems. Solutions containing silicon as six different compounds in xylene were prepared and analyzed through ICP-AES. At low liquid flow rates, the h-TISIS improves the sensitivity and the limits of detection as compared to the d-DIHEN. In contrast, shorter wash-out times were obtained for the d-DIHEN. As regards to the effect of the silicon chem. form, h-TISIS and d-DIHEN have minimized it with respect to a conventional cyclonic spray chamber.
