Photocatalytic oxidation of tetracycline by magnetic carbon-supported TiO2 nanoparticles catalyzed peroxydisulfate: Performance, synergy and reaction mechanism studies

TiO2 decorated on ferroferric oxides coupled with activated carbon (TFOC), as a magnetic heterogeneous catalyst, was synthesized and its photocatalytic ability was assessed in coupling with UV light towards peroxydisulfate (PS) activation to degrade tetracycline (TC). Main properties of the catalyst were analyzed by using UV-visible DRS, FESEM, VSM, PL, EDS, TEM, XRD and BET techniques. A proposed reaction mechanism was suggested for activating PS and TC degradation. Both TiO2 and Fe3O4 (FO) were synthesized and dispersed successfully on the carbon surfaces in nano-sized scale. It was found that the photocatalytic activity of TiO2 was accelerated significantly after loading on the magnetic carbon. TFOC showed an excellent magnetic response in the presence of a magnet. According to PL analysis, FO nanoparticles and carbon played a critical role in the suppression of recombination of excited electrons and holes. TFOC in coupling with UV showed a best performance, compared to the other single and binary processes for TC decontamination. After 15 min reaction, the highest level of elimination, 96% TC (60 mg/L) and 72.1% TOC were obtained at optimum operational conditions: pH:5.0, TFOC: 0.3 g/L and PS:1.0 mM. A noticeable synergistic effect was found for TFOC in combination with UV and PS. Both negligible Fe leaching and high efficiency of degradation were achieved with reused times, illustrating that TFOC has an excellent durability and reusability. Based on trapping tests, the importance of involved oxidizing species during TC decontamination over TFOC/PS/UV system was in order of O-1(2) > SO4 center dot- > h(+) > O-2(center dot-) > HO center dot. Among studied anions, the highest quenching impact was observed for chloride ions. Particularly, TFOC composite showed the high catalytic activity as well excellent potential in recovery and its coupling with UV light and PS can be introduced as a promising strategy for efficient destroying TC antibiotic.

Hydrogels with multifunctional properties such as biocompatibility, adhesion, and self-healing and inherent antibacterial properties are of great significance in biomedical applications, especially for wound dressings and drug release. However, it remains a significant challenge to combine all of these properties in a one-component hydrogel system. Furthermore, most injectable hydrogels are mechanically weak and not self-healing, which limits their applications as wound dressings. Herein, we designed a hydrogel with a tunable two-layer structure that endows the hydrogel with adhesion, self-healing, and antibacterial properties in different layers. First, chitosan (CHI) was oxidized to aldehyde-chitosan (ACHI). The Schiff base reaction was then used to fabricate the CHI/ACHI hydrogel resulting in the first layer. For the second layer, a physical cross-linking process using phytic acid (PA) as a sequential cross-linker was achieved, resulting in a CHI/ACHI/PA hydrogel. The CHI/ACHI hydrogel exhibited good injectable, adhesion, and self-healing properties. After cross-linking with PA, the CHI/ACHI/PA hydrogel showed enhanced mechanical properties, which enabled it to act as a protective layer. Additionally, the antibacterial test and cytotoxicity tests performed on this two-layer structured CHI/ACHI hydrogel exhibited excellent inherent antibacterial activity and good biocompatibility. The biocompatible injectable hydrogel was endowed with more possibilities relying on its self-healing and antibacterial abilities. These properties endorse the CHI/ACHI two-layer hydrogel as an excellent candidate for biological applications, especially wound dressings.

Human extrahepatic and intrahepatic cholangiocyte organoids show region-specific differentiation potential and model cystic fibrosis-related bile duct disease

The development, homeostasis, and repair of intrahepatic and extrahepatic bile ducts are thought to involve distinct mechanisms including proliferation and maturation of cholangiocyte and progenitor cells. This study aimed to characterize human extrahepatic cholangiocyte organoids (ECO) using canonical Wnt-stimulated culture medium previously developed for intrahepatic cholangiocyte organoids (ICO). Paired ECO and ICO were derived from common bile duct and liver tissue, respectively. Characterization showed both organoid types were highly similar, though some differences in size and gene expression were observed. Both ECO and ICO have cholangiocyte fate differentiation capacity. However, unlike ICO, ECO lack the potential for differentiation towards a hepatocyte-like fate. Importantly, ECO derived from a cystic fibrosis patient showed no CFTR channel activity but normal chloride channel and MDR1 transporter activity. In conclusion, this study shows that ECO and ICO have distinct lineage fate and that ECO provide a competent model to study extrahepatic bile duct diseases like cystic fibrosis.

Ethnopharmacological relevance: Minthostachys verticillata (Griseb.) Epling (Lamiaceae), known as Peperina is a medicinal native plant, with a traditional use as a digestive, antispasmodic and antidiarrheic. Aim Of The Study: Despite its folkloric use, no scientific evaluation of this plant related to the gastrointestinal inflammatory process has been carried out to date. The present study aims to assess the effects of M. verticillata on gastrointestinal system in experimental models. Materials and methods: M. verticillata (250 and 500 mg/kg) was orally tested in a colitis model induced by acetic acid. Colon weight/length ratio, oxidative stress (oxidized and reduced glutathione), histological changes using Alcian blue and hematoxylin & eosin staining and expression of IL1 beta, TNF alpha, iNOS, COX-2 were evaluated. The effect of the extract in three additional in vivo models were studied: intestinal motility and diarrhea induced by ricin oil, and visceral pain induced by intracolonic administration of capsaicin. Finally, the activity on concentration response curves of acetylcholine, calcium chloride, potassium and serotonin were achieved in isolated rat jejunum. Results: In the colitis model, M. verticillata induced a significant reduction in the colon weight/length ratio, oxidative stress and expression levels of IL-1 beta, iNOS and COX-2. Also, the extract diminished the severity of microscopic tissue damage and showed protective effect on goblet cells. Intestinal motility, diarrhea, visceral pain-related behaviors and referred hyperalgesia were significantly reduced when the animals were treated with the extract. Furthermore, in isolated jejunum, M. verticillata significantly reduced the contraction induced by serotonin and acetylcholine. Likewise, the extract non-competitively inhibited the response-concentration induced by CaCl2 and inhibited both low and high K+-induced contractions. Conclusions: This is the first study to validate traditional use of M. verticillata for digestive disorders and demonstrated that its aqueous extract could represent a promising strategy in targeting the multifactorial pathophysiology of inflammatory bowel disease.

Adult male Wistar rats were randomly divided into four groups: (a) control (normal saline, orally), (b) cadmium chloride (0.1 mg/kg, single dose, intraperitoneally), (c) Feijoa fruit extract (400 mg/kg, orally for 30 consecutive days) and (d) CdCl2 + Feijoa fruit extract. One day after receiving the last medicine, the LH, FSH, prolactin and testosterone concentration were assayed. Also, sperm parameters and tissue structure of the testis were evaluated. Administration of Feijoa fruit extract after CdCl2 injection in rats ameliorated sperm parameters such as sperm count, morphology, motility and sperm viability, increased levels of LH, FSH, prolactin and testosterone and improved testicular histology. According to the results of this study, it was shown that Feijoa can reduce the destructive side effects of CdCl2 on testicular tissue and sex hormones of the pituitary-gonadal pathway.
