Effectiveness of a formulation containing peptides and vitamin C in treating signs of facial ageing: three clinical studies

Objective Vitamin C and peptides are widely used in cosmetic products but there is a paucity of clinical studies showing that the formulations are effective in treating signs of facial ageing. These 3 clinical studies evaluated the effectiveness of an anti-ageing formula containing natural vitamin C (10%), biopeptides (rice and lupin), hyaluronic acid, and Vichy volcanic mineralising water, in amber glass ampoules with no preservatives (Peptide-C ampoules). Methods Dansyl chloride fluorescence labelling compared cell turnover for Peptide-C ampoules vs untreated skin in 32 female subjects. Study 2, an open clinical study, evaluated the efficacy on wrinkles of Peptide-C ampoules by investigator clinical scoring based on Dynamical Atlas visual assessment (N = 40) and subject self-assessment questionnaires (N = 47). Study 3, an open clinical study, evaluated wrinkles by instrumental quantification with 3D fringe projection analysis (N = 40) and subject questionnaires (N = 51). Results The mean cell turnover was faster for skin treated with Peptide-C ampoules compared to untreated skin (17.1 days vs. 19.2 days; P < 0.0001). In study 2, after 28 days application of Peptide-C ampoules, clinical grading of crow's-feet wrinkles, forehead wrinkles and nasolabial folds decreased by 9%, 11% and 5%, respectively (all P < 0.05 vs baseline). Anesthetic potential of the essential oils of Aloysia triphylla, Lippia sidoides and Mentha piperita for Colossoma macropomum

The aim of this study was to evaluate the anesthetic potential of the essential oils of Aloysia triphylla (EOAT), Lippia sidoides (EOLS) and Mentha piperita (EOMP) in relation to juvenile tambaqui (Colossoma macropomum), based the swimming behavior of these fish and possible histopathological damage to the gills. To induce anesthesia, juvenile tambaqui (130.55 +/- 22.4 g; 16.8 +/- 0.95 cm) were individually exposed to different concentrations of the essential oils (EOAT: 5, 10, 20, 40, 60, 80 and 150 mg L-1; EOLS: 2, 5, 10, 20, 30, 40 and 50 mg L-1; EOMP: 5, 10, 20, 40, 60, 70 and 90 mg L-1), for exposure periods of between four minutes and six hours. The three essential oils evaluated presented anesthetic properties. The concentration intervals over which the fish reached all the stages of anesthesia (1, 2, 3a, 3b and 4) were between 40 and 150 mg L-1 for EOAT; 20 and 50 mg L-1 for EOLS; and 40 and 90 mg L-1 for EOMP. The times taken to reach the anesthesia stages were inversely proportional to the increases in the concentration of the essential oils. The histopathological damage to the gills of the tambaqui that resulted from using essential oil concentrations that were defined as anesthetic (150 mg L-1 EOAT, 30 mg L-1 EOLS and 90 mg L-1 EOMP) were also evaluated. The main damage recorded was hypertrophy and hyperplasia of the lamellar epithelium, lamellar fusion and proliferation of chloride cells. According to the histopathological alteration index (HAI), there were no irreversible damage (such as fibrosis and necrosis) to the gills of the tambaqui. However, while EOLS and EOMP caused mild damages in the gills, the EOAT generated more severe damages. Overall, for the tambaqui, the essential oils of A. triphylla, L. sidoides and M. piperita are efficient anesthetic in the doses of 150, 30 and 90 mg L-1, respectively.

Antibiotics, algal evaluations and subacute effects of abattoir wastewater on liver function enzymes, genetic and haematologic biomarkers in the freshwater fish, Clarias gariepinus

Abattoirs are positioned close to freshwaters in most developing countries where untreated wastewaters are discharged into with potential risk to aquatic life such as fish and ultimately human health. We assessed physicochemical parameters, antibiotics (oxytetracycline and diclofenac) and algal load of effluent collected from a major abattoir in Nigeria. Furthermore, liver function enzymes, genotoxic and haemotoxic effects of subacute concentration (10% of 96 hLC(50) value) of the wastewater were evaluated over a period of 28 d in Clarias gariepinus (The African Sharptooth Catfish). The 96 hLC(50) value of the abattoir wastewater against C. gariepinus was 154.14 mL/L (15.4%). Nitrates, phosphates, sulphates, chloride, ammonia, TDS, TSS, BOD5 and heavy metals (Fe and Pb) in the wastewater were above permissible limits while diclofenac and oxytetracycline were below detection limit (BDL). Microalgae in the wastewater were mostly Bacilliariophyta (Navicula spp.) (45.64%) and euglenoids (Euglena and Phacus spp.) (49.48%). Liver function enzymes (LDH, AST, ALT) level were higher in exposed fishes except for ALP which was lower at day 28 compared to control levels. Erythrocytic genotoxic indices (nuclear abnormalities) were significantly higher (p < 0.05) in the exposed fishes particularly at day 28 compared to control. Haematologic indices level such as WBC, MCV MCH, MCHC increased significantly (p < 0.05) while lymphocytes, HGB, RBC, HCT levels decreased significantly (p < 0.05) in the exposed C. gariepinus by day 28 compared to control. These results demonstrate potential adverse effects posed to aquatic fish species in the Ogun River by the discharge of the abattoir effluent. The microalgae species identified in the effluent may be explored for pre-treatment of the effluent before discharge in order to prevent eutrophication and increased pollutant load in the River. The study results will contribute to evidence-based environmental risk management of the River which is relevant to the UN SDGs 6 (clean water and sanitation), 11 (sustainable cities and communities) and 14 (sustaining life below water). Uranium(VI) toxicity in tobacco BY-2 cell suspension culture – A physiological study

For the first time, the physiological and cellular responses of Nicotiana tabacum (BY-2) cells to uranium (U) as an abiotic stressor were studied using a multi-analytic approach that combined biochemical analysis, thermodynamic modeling and spectroscopic studies. The goal of this investigation was to determine the U threshold toxicity in tobacco BY-2 cells, the influence of U on the homeostasis of micro-macro essential nutrients, as well as the effect of Fe starvation on U bioassociation in cultured BY-2 cells. Our findings demonstrated that U interferes with the homeostasis of essential elements. The interaction of U with BY-2 cells confirmed both timeand concentration-dependent kinetics. Under Fe deficiency, a reduced level of U was detected in the cells compared to Fe-sufficient conditions. Interestingly, blocking the Ca channels with gadolinium chloride caused a decrease in U concentration in the BY-2 cells. Spectroscopic studies evidenced changes in the U speciation in the culture media with increasing exposure time under both Fe-sufficient and deficient conditions, leading us to conclude that different stress response reactions are related to Fe metabolism. Moreover, it is suggested that U toxicity in BY-2 cells is highly dependent on the existence of other micro-macro elements as shown by negative synergistic effects of U and Fe on cell viability.

Anoctamin 1 antagonism potentiates conventional tocolytic-mediated relaxation of pregnant human uterine smooth muscle

Background Currently available tocolytic agents are not effective treatment for preterm labor beyond 48 h. A major reason is the development of maternal side effects which preclude the maintenance of an effective steady-state drug concentration. One strategy that can mitigate these side effects is utilizing synergistic drug combinations to reduce the drug concentrations necessary to elicit a clinical effect. We have previously shown that three anoctamin 1 (ANO1) antagonists mediate potent relaxation of precontracted human uterine smooth muscle (USM). In this study, we aimed to determine whether a combination of sub-relaxatory doses of tocolytic drugs in current clinical use [the L-type voltage-gated calcium channel (VGCC) blocker, nifedipine (NIF); and the beta(2)-adrenergic (beta 2AR) agonist, terbutaline (TRB)] will potentiate USM relaxation with two ANO1 antagonists [benzbromarone (BB) and MONNA (MN)]. Objective This study sought to examine the synergistic potency and mechanistic basis of two ANO1 antagonists with currently available tocolytic drugs. Functional endpoints assessed included relaxation of pre-contracting pregnant human USM tissue, inhibition of intracellular calcium release, and reduction of spontaneous transient inward current (STIC) recordings in human uterine smooth muscle cells. Methods Human myometrial strips and primary human USM cells were used in organ bath and calcium flux experiments with different combinations of sub-threshold doses of ANO1 antagonists and terbutaline or nifedipine to determine if ANO1 antagonists potentiate tocolytic drugs. Results The combination of sub-threshold doses of two ANO1 antagonists and current tocolytic drugs demonstrate a significant degree of synergy to relax human pregnant USM compared to the effects achieved when these drugs are administered individually. Conclusion A combination of sub-threshold doses of VGCC blocker and beta 2AR agonist with ANO1 antagonists potentiates relaxation of oxytocin-induced contractility and calcium flux in human USM ex vivo. Flame retardancy of particleboards made from oil palm trunk-poly(vinyl) alcohol with citric acid and calcium carbonate as additives

Flame retardant particleboards from oil palm trunk particles were made by treatment with sodium chloride solution, then bonded with poly(vinyl) alcohol together with citric acid and calcium carbonate as additives. Flame retardancy, thermal, physical, and mechanical properties were evaluated for suitability as construction materials. The novel flame-retardant oil palm trunk particleboard was successfully produced. Treated oil palm trunk with 20% of poly(vinyl) alcohol and 10% of calcium carbonate showed the highest limited oxygen index (LOI) with good thermal stability and potential to be used as flame-retardant particleboard. Succinyl chitosan gold nanocomposite: Preparation, characterization, in vitro and in vivo anticandidal activity

Herein, we have successfully synthesized a novel N-Succinyl chitosan/gold nanocomposite (N-SuC/Au NC) using N-SuC and gold(III) chloride, and investigated the biocompatibility and antifungal activity. The synthesized N-SuC/Au NC was characterized by UV-visible spectroscopy, X-ray diffraction, field emission scanning electron microscope, and inductively coupled plasma atomic emission spectroscopy. The N-SuC/Au NC exhibited a strong inhibition effect towards pathogenic Candida albicans. Morphological analysis revealed the destruction of C. albicans cell membrane due to N-SuC/Au NC treatment. The in vitro and in vivo toxicity of N-SuC/Au NC was analyzed with HEK293T mammalian cells and zebrafish larvae, respectively. The synthesized N-SuC/Au NC demonstrated no cytotoxicity towards HEK293T cells up to 1200 mu g/mL concentration. The survival rate of the zebrafish larvae at 120 hpf, was found as 100% up to 1200 mu g/mL of N-SuC/Au NC exposure. Activation of potassium peroxymonosulfate for rhodamine B photocatalytic degradation over visible-light-driven conjugated polyvinyl chloride/Bi2O3 hybrid structure

Survival of MS2 and phi 6 viruses in droplets as a function of relative humidity, pH, and salt, protein, and surfactant concentrations

The survival of viruses in droplets is known to depend on droplets’ chemical composition, which may vary in respiratory fluid between individuals and over the course of disease. This relationship is also important for understanding the persistence of viruses in droplets generated from wastewater, freshwater, and seawater. We investigated the effects of salt (0, 1, and 35 g/L), protein (0, 100, and 1000 mu g/mL), surfactant (0, 1, and 10 mu g/mL), and droplet pH (4.0, 7.0, and 10.0) on the viability of viruses in 1-mu L droplets pipetted onto polystyrene surfaces and exposed to 20%, 50%, and 80% relative humidity (RH) using a culture-based approach. Results showed that viability of MS2, a non-enveloped virus, was generally higher than that of phi 6, an enveloped virus, in droplets after 1 hour. The chemical composition of droplets greatly influenced virus viability. Specifically, the survival of MS2 was similar in droplets at different pH values, but the viability of phi 6 was significantly reduced in acidic and basic droplets compared to neutral ones. The presence of bovine serum albumin protected both MS2 and phi 6 from inactivation in droplets. The effects of sodium chloride and the surfactant sodium dodecyl sulfate varied by virus type and RH. Meanwhile, RH affected the viability of viruses as shown previously: viability was lowest at intermediate to high RH. The results demonstrate that the viability of viruses is determined by the chemical composition of carrier droplets, especially pH and protein content, and environmental factors. These findings emphasize the importance of understanding the chemical composition of carrier droplets in order to predict the persistence of viruses contained in them.
