Cataracts have a high incidence and prevalence rate worldwide, and they are the leading cause of blindness. Lens epithelial cell (LEC) apoptosis is often analysed in cataract research since it is the pathological basis of cataracts, except for congenital cataract. Chloride channels are present in ocular tissues, such as in trabecular cells, LECs and other cells. They serve an important role in apoptosis and participate in endoplasmic reticulum (ER) stress and oxidative stress. However, their role in the apoptosis of LECs has not been discussed. The present study examined the effects of the chloride channel blocker 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) in human LECs (HLECs) to elucidate the role of NPPB in HLECs and investigate the role and mechanism of chloride channels in cataract formation. HLECs were exposed to NPPB. Cell survival rate was evaluated using Cell Counting Kit-8 assays. Oxidative stress was detected as reactive oxygen species (ROS) in cells by using a ROS assay kit. Apoptosis was examined by assessing mitochondrial membrane potential and using a JC-1 assay kit, and western blot analysis was performed to measure the expression levels of mitochondrial-dependent apoptosis pathway-associated proteins. ER stress was evaluated by determining the intracellular calcium ion fluorescence intensity, and western blot analysis was performed to measure ER stress-associated protein expression. The results revealed that NPPB treatment decreased the viability of HLECs and increased apoptosis. Additionally, NPPB increased intracellular ROS levels, as well as the number of JC-1 monomers and the protein expression levels of B-cell lymphoma-2 (Bcl-2)-associated X and cleaved caspase-3, and decreased Bcl-2 protein expression. NPPB increased intracellular calcium ions, the protein expression levels of activating transcription factor 6, JNK, C/EBP homologous protein and caspase-12, and the phosphorylation of protein kinase R-like endoplasmic reticulum kinase. N-acetylcysteine and 4-phenylbutyric acid inhibited NPPB-induced oxidative stress, ER stress and apoptosis. Therefore, NPPB treatment decreased cell viability and promoted apoptosis of HLECs via the promotion of oxidative and ER stress.

Landfill refuse is a mixture of inorganic minerals and organic matter that is capable of undergoing complexation and redox reactions due to its active functional groups. Organic matter often combines with minerals in landfill refuse and it remains unclear whether this combination involves electron transfer. Therefore, the effects of landfill refuse composition on reductive dechlorination and speciation transformation of heavy metals were investigated in this study. Results show that landfill refuse comprises protein- and humic-like substances, aliphatic structures, and a large number of hydroxyl, carboxyl, quinoid and other active functional group. The electron donating capacity (0.09-0.26 mu mol/g(C)) of landfill refuse was found to be higher than its electron accepting capacity (0.03-0.23 mu mol/g(C)), indicating that electron donating groups (hydroxyl) were the main redox-active moieties, facilitating the reductive dechlorination of pentachlorophenol (PCP) by microorganism. Fe2O3, FeO and SiO2 were the main inorganic minerals affecting PCP dechlorination. The speciation distribution of heavy metals in landfill refuse was determined by the BCR sequential extraction method. Results showed that Zn and Ni have high potential migration capacity, poor stability and the highest bioavailability, while Cr, Cu and Pb are relatively stable and have weak migration potential. The oxygen- and nitrogen-containing functional groups, aliphatic structures and aromatic carbon in landfill refuse can promote the transformation of Ni and Cr from an unstable to stable state. Protein-like substances exhibit a strong Cu binding ability, allowing Cu to combine with organic matter more easily than other assessed heavy metals. Both Fe2O3 and FeO affected the stability of Cu. FeO promoted the stabilization of Zn, whereas Fe2O3 and SiO2 promoted Cu instability. This paper discusses the corrosion behavior of stirrups in two concrete beams under sustained loading, exposed to a chloride environment for 9 years. The cracking maps of concrete beams were recorded after 28 days and after 9 years, respectively. The corrosion maps of the stirrups were drawn and the cross-sectional losses of stirrups were measured after destroying the beams. The experimental results show that the horizontal legs of stirrups mainly corroded at the mid span of the tensile zone and at the ends of the compressive zone of the reinforced concrete beams regardless of the exposure conditions, while the opposite horizontal legs were protected. The corrosion of vertical legs of stirrups, which was slight, was not impacted by the load-induced cracks. In the present study, it is difficult to find the correlation of corrosion degree between stirrups and the main reinforcing steels in the reinforced concrete beams. The corrosion behavior of the intersection between stirrups and main bars was controlled by the interfacial conditions between them. Cathodic protection occurred at the ends of the top-cast side and the mid-span of bottom-cast side of reinforced concrete beams when the compressive rebars and tensile rebars were placed at the top- and bottom- cast sides, respectively. A fabric with different functions on its two sides has unparalleled advantages for practical use. However, it is difficult to graft different functional polymers onto the two sides of cotton fabric by conventional modification methods. Herein, we report a novel approach based on a combination of grafting through methodology and the mist polymerization technique to achieve this objective. Acrylic acid (AA) was first linked onto cotton fibers via an esterification reaction. The incorporated AA moieties on the fiber surfaces were further copolymerized with other monomers using the mist polymerization technique to graft different functional coatings. Poly(methacryloxyethyltrimethyl ammonium chloride) was grafted on one side of the cotton fabric, and the opposite surface was modified by the polymerization of AA followed by the esterification of polyethylene glycol-400 (PEG-400). As a result, two functional coatings were constructed on the two sides of the fabric, i.e., antibacterial and antifouling coatings. Skin stimulation tests and toxicity assessments demonstrated that the resulting fabric is safe for skin contact. Moreover, the desired cotton properties were not significantly reduced by the new surface modification process.

Methods Oral squamous cell carcinoma cells (CAL27) were incubated with increasing IS doses and then submitted to proliferation (2D), cell death (2D) and spheroid assay (3D). Results The IS anti-tumor effect was dependent on both its N-alkyl chain length and anion, whereby C(16)MImCl proved to be more effective in combination for inhibiting cell proliferation and cell-cell adhesion, outperforming the methylated C(16)M(2)ImCl derivative and, most importantly, the gold standard-cisplatin. In addition, C(16)MImCl had little effect on keratinocytes and more pronounced effects on more aggressive tumor cells. It also exhibited similar effects on inducing cell death when compared to Cisplatin. This compound spread to a greater area of the tumor sphere and produced an enhanced number of apoptotic and necrotic cells in the tumor cell line, demonstrating only a small rise in the healthy cells. Conclusion These data indicate that the effect of C(16)MlmCl on OSCC is promising, as it is selective for cancer cells.

BACKGROUND The cys-loop ligand-gated ion channels, including the glutamate-gated chloride channel (GluCl) and GABA-gated chloride channel (Rdl) are important targets for drugs and pesticides. Recently, an additional I321T mutation in GluCl3 was identified in several abamectin resistant T. urticae field populations. Here, we aim to functionally validate this mutation and determine its phenotypic strength. RESULTS The GluCl3 I321T mutation was introgressed into a T. urticae susceptible background by marker-assisted backcrossing, revealing contrasting results in phenotypic strength, ranging from almost none to 50-fold. Next, we used CRISPR-Cas9 to introduce I321T, G314D and G326E in the orthologous Drosophila GluCl. Genome modified flies expressing GluCl I321T were threefold less susceptible to abamectin, while CRISPRed GluCl G314D and G326E flies were lethal. Last, functional analysis in Xenopus oocytes revealed that the I321T mutation might reduce GluCl3 sensitivity to abamectin, but also suggested that all three T. urticae Rdls are affected by abamectin. CONCLUSION Three different techniques were used to characterize the role of I321T in GluCl3 in abamectin resistance and, combining all results, our analysis suggests that the I321T mutation has a complex role in abamectin resistance. Given the reported subtle effect, additional synergistic factors in resistance warrant more investigation.

In addition, the association constant (K-M) and the kinetics of the template reaction showed that the MW-TP was confirmed to be free radical initiated polymerization and I Zip-up (ZIP) polymerization mechanism, which demonstrated the formation of the cationic fragment structure again. This new cationic fragment structure greatly enhanced the ability of charge neutralization, electrical patching and adsorption bridging, thus improving the flocculation performance. Compared with cationic P(AM-DMC) (CPADM), Commercial cationic polypropanamide (CCPAM-1 and CCPAM-2), TPADM had better flocculation effect on coal washing wastewater. Under the optimum flocculation conditions (dosage = 16 mg/L, pH = 7.5), suspended solid (SS) concentration and turbidity of coal washing wastewater treated by TPADM reached 14.3 mg/L and 8.7 NTU, respectively. The floc formation, breakage, and regeneration analyses showed that the cationic fragment structure in TPADM could contribute to the formation of large and high density flocculation structure (d(50) >= 245 mu m,D-f >= 2.05), and these coal flocs can regenerate rapidly after being broken. Finally, these large and compact flocs help to accelerate the flocs sedimentation and reduce the SS concentration and turbidity of coal washing wastewater, and enhance the effect of solid-liquid separation.

A higher P(3HB) content of 0.42 g g(-1) was achieved when nitrogen was limited compared to 0.34 g g(-1) under phosphate-limiting conditions; however, sucrose utilization was better when phosphate was limited. Replacing NaCl by KCl and evaluating different concentrations ranging from 0.08 to 1.7 mol L-1 in the process medium showed that B. megaterium has a higher tolerance to KCl as the biomass and P (3HB) formation was increased to 0.48 g g(-1) compared to 0.36 g g(-1). The combination of applying KCl instead of NaCl together with phosphorous limitation significantly increased P(3HB) productivity to 0.25 g L-1 h(-1) compared to 0.09 g L-1 h(-1).
