The authors describe the synthesis of atipamezole hydrochloride starting from the reaction of 1,2-di(bromomethyl)benzene and 2,4-pentanedione in the presence of NaOH under phase transfer catalytic conditions, followed by alkylation with EtI to obtain 2-acetyl-2-ethylindan. Next, 2-acetyl-2-ethylindan is coupled with aminoethanol to afford a ketimine, which is, then, brominated. The resulting 2-(bromoacetyl)-2-ethylindan is coupled with excess formamide, followed by treatment with HCl in MeOH to give atipamezole hydrochloride.
