ERK1 and ERK5 are proposed to have pivotal roles in several types of cancer. Under some circumstance, ERK5 may provide a common bypass route, which rescues proliferation upon abrogation of ERK1 signaling. Thus, we accurately classified the tumor types from The Cancer Genome Atlas (TCGA) based on the expression levels of ERK1 and ERK5. We proposed a novel therapeutic strategy to overcome the above-mentioned compensatory mechanism in specific tumor types by co-targeting both ERK1 and ERK5. On the basis of the idea of overcoming ERK5 compensation mechanism, 22ac (ADTL-EI1712) as the first selective dual-target inhibitor of ERK1 and ERK5 was discovered to have potent antitumor effects in vitro and in vivo. Interestingly, this compound was found to induce regulated cell death accompanied by autophagy in MKN-74 cells. Taken together, our results warrant the potential of this dual-target inhibitor as a new candidate drug that conquers compensatory mechanism in certain tumor types.

In this work, we applied a novel dithiocarbamate-grafted star-like polymer for efficient reclamation of cationic dyes. This material is highly selective towards cationic dyes and almost completely rejective to anionic dyes. The adsorption kinetics, isotherms, and thermodn. were systematically investigated to analyze the adsorption behavior of cationic dyes onto the material. As compared with other reported adsorbents, this cheap and facilely-prepared material has exceptional adsorption capacities for industrial cationic dyes, e.g., methylene blue (MB, 2624 mg/g), crystal violet (CV, 2553 mg/g), basic fuchsin (BF, 1729 mg/g), and methylene violet 3RAX (MV, 1239 mg/g), being 5-11 times of that reported in literatures. Based on the effects of pH and ionic strength on adsorption, competitive adsorption of the cationic dyes, and FTIR characterization, mechanisms for the adsorption and desorption of cationic dyes were proposed. The adsorbed dyes can be recovered over 95% just by immersing into diluted HCl solution (0.01 M) for 10 min, thus avoiding organic solvent extraction Over 98% of the adsorption capacity was maintained during consecutive reclamation operation. Due to the high selectivity, high adsorption capacity and the ease of efficient regeneration, this material is potentially applicable for targeted reclamation of cationic dyes from dyeing wastewater.

A series of novel anthranilic diamides analogs I [R = 4-F, 5-F, 5-Cl, 5-Br, 4-F3C; R1 = Me, Et, i-Pr, etc.] containing trifluoroethoxyl pyrazole moiety was designed and synthesized and their insecticidal bioactivities against Mythimna separata (Walker, M. separata) and Plutellaxylostella (P. xylostella) were evaluated. The structures of the title compounds I were confirmed by 1H NMR, 13C NMR and HRMS. Preliminary insecticidal activities showed that some of the title compounds possessed good to excellent bioactivities towards M. separata and P. xylostella. Compounds I [R = 5-F, R1 = i-Pr; R = 4-F, R1 = F3CH2C] exhibited 100% mortality rate against M.sep. at 0.2mg/L. For the P. xylostella, the synthesized compoundsI [R = 5-F, R1 = i-Pr, cyclopropyl; R = 5-F3C, 5-Br, R1 = Me; R = 5-Cl, R1 = HF2CH2C] showed 70%, 80%, 75%, 65% and 60% insecticidal activities at 1×10-6 mg/L, resp., higher than that of chlorantraniliprole(0). Based on excellent insecticidal activities, the mode of the action was tested by the calcium-imaging technique, the results of which demonstrated that the novel compounds shared the same target with chlorantraniliprole. The binding pose of the most active compound I [R = 4-F, R1 = F3CH2C] in RyRs of P. xylostella was predicted by mol. docking, which showed that compound I [R = 4-F, R1 = F3CH2C] interacted with the residues Glu140(A) and His147(A) via hydrogen bond.

The process represents a new addition to the limited asym. reactions of isobenzopyryliums, a family of versatile 10π-electron aromatic species. Excellent asym. induction is achieved for the first time without an anchoring group in the 4-position or a metal catalyst, both of which were required previously in these reactions. The success is attributed to the unusual chiral counteranion (meanwhile also the nucleophile) generated in situ from the chiral phosphate and the boronic acid as well as the leaving group. Preliminary control experiments provided important insight into the reaction mechanism.

Mono-(2-O-tosyl)-β-cyclodextrin (β-CD-2-OTs) was produced under a chem. reaction between β-cyclodextrin and TsCl with a mole ratio of 1:6 in alk. aqueous solution 2-ethylenediamio-β-cyclodextrin (β-CD-2-E) was synthesized under the reaction between β-CD-2-OTs and ethylenediamine at the temperature around 60 °C. Mono-(2-(2,3-dibromopropionyl)ethylenediamino-2-deoxy)-β-cyclodextrin was synthesized through the reaction between β-CD-2-E and 2,3-dibromopropionyl chloride. The synthesis process was proved to be successful through the anal. of FT-IR spectra and 1H-NMR spectrum. The novel finishing agent was proved to graft onto silk successfully through the SEM pictures.

α-Bromoacrylamidoethyl cyclodextrin (β-CD-2-EDBA) was prepared with β-cyclodextrin, p-toluenesulfonyl chloride and 2,3-dibromo-propionyl chloride. Aroma finishing of silk fabric grafted with 8-CD-2-EDBA was carried out. Optimum grafting process was pH value 9, grafting agent 16% (owf), sodium sulfate 8 g/L, liquor ratio 1:50, and treating at 90°C for 40 min. Grafting ratio of treated silk fabric was high, and fragrance storage and fragrance retention time enhanced significantly. Scanning electron microscope picture proved that B-CD-2-EDBA was successfully grafted onto silk surface. Grafting ratio was tested with phenolphthalein spectral probe method and gravimetric method sep., it was found that the former had higher grafting ratio, while the latter was more accurate.
