Kinetics of free radical polymerization of Me methacrylate using potassium peroxomonosulfate as initiator in the presence of benzyltributylammonium chloride (BTBAC) as phase transfer catalyst was studied. The polymerization reactions were carried out under nitrogen atm. and unstirred conditions at a constant temperature of 60°C in Et acetate/water bi-phase system. The role of concentrations of monomer, initiator, catalyst, temperature, acid and ionic strength on the rate of polymerization (Rp) was ascertained. The orders with respect to monomer, initiator and phase transfer catalyst were found to be 1.5, 0.5 and 0.5 resp. The rate of polymerization (Rp) is independent of ionic strength and pH. Based on the kinetic results, a suitable mechanism is proposed.

Conditions of glass capillary (50 μm inner diameter) inner surface modification by an aqueous solution of ammonia were studied. The resulting silica gel porous layer was modified chem. by methyloctyldichlorosilane. Under given etching conditions (temperature 250-300° for 16 h), a 5-10 volume% aqueous solution of ammonia was optimal. The static way of silylation by 10 volume% silane solution in toluene for 16 h at 250-280° with the column filled to 50-70% of its volume is optimal. This method provides a mech. stable porous layer 1-10 μm thick.

A series of 7β-[(Z)-2-(2-aminothiazol-4-yl)-2-hydroxyiminoacetamido]-3-(heteroarylmethylthio)cephalosporins was designed, synthesized and evaluated for antibacterial activity and oral absorption in rats. Antibacterial activity was markedly influenced by the structure of the heteroaromatic ring moiety. Oral absorption was influenced by the heteroaromatic ring moiety as well as by the arrangement of heteroatoms. Among these compounds, FK041, 7β-[(Z)-2-(2-aminothiazol-4-yl)-2-(hydroxyimino)acetamido]-3-[(pyrazol-3-yl)methylthio]-3-cephem-4-carboxylic acid, showed potent antibacterial activity against both Gram-pos. and Gram-neg. bacteria including Haemophilus influenzae. Further, it showed higher oral absorption than CFDN.

Sodium glucose co-transporter 1 (SGLT1) plays a dominant role in the absorption of glucose in the gut and is considered a promising target in the development of therapeutic options for postprandial hyperglycemia. Previously, we reported potent and selective SGLT1 inhibitors 1 and 2 showing efficacy in oral carbohydrate tolerance tests in diabetic rat models. In a pharmacokinetic (PK) study of glycoside I (R = OH), excessive systemic exposure to metabolites of I (R = OH) was observed, presumably due to the high permeability of its aglycon. To further improve SGLT1 inhibitory activity and reduce aglycon permeability, a series of 4-benzyl-5-isopropyl-1H-pyrazol-3-yl β-D-glycopyranoside derivatives, e.g. I (R = NH2), bearing novel hydrophilic substitution groups on the Ph ring were synthesized and their inhibitory activity toward SGLTs was evaluated. Optimized compound II showed an improved profile satisfying both higher activity and lower permeability of its aglycon. Moreover, the superior efficacy of II in various carbohydrate tolerance tests in diabetic rat models was confirmed compared with acarbose, an α-glucosidase inhibitor (α-GI) widely used in the clinic.

Diethylamino-substituted diarylethenes can switch between P-type photochromism (P-type = thermally stable isomers) and T-type (thermally unstable isomers) photochromism upon addition of trifluoromethanesulfonic acid. The protonated closed-form photoinduced color isomers return to open-ring isomers at a fast rate at elevated temperature such as 160° C. The protonation and deprotonation of the open- and closed isomers was reversible. Such photochromic materials could be used as reversible switching materials for write-by-light/erase-by-heat recording.

The synthesis, characterization, and exploratory chem. of two classes of alkene-platinum-silyl complexes, which have been postulated as hydrosilation intermediates, are described in this report. The unique dimeric complexes 1, [R3Si(μ-Cl)(η2-COD)Pt]2 {R3Si = Et3Si, MeCl2Si, Me2ClSi, “(EtO)3Si”, PhMe2Si, and (Me3SiO)Me2Si; COD = cycloocta-1,5-diene}, and the bis-silyl complexes 2, (η4-COD)Pt(SiR3)2 (R3Si = Cl3Si, MeCl2Si, Me2ClSi, PhMe2Si), are formed from the sequential reaction of 2 and 4 equivalent of the corresponding hydrosilanes, resp., with Pt(COD)Cl2 in the presence of a small excess of COD. Complexes 1 are stable for many days in solution at room temperature but decompose via slow elimination of chlorosilane. Some of the bis-silyl compounds 2 are stable for extended periods under inert atm. and especially below 0°, either in the solid state or in solution (in the presence of a small excess of free COD). Complexes 2 display catalytic activity as discrete, mol., and mononuclear species for hydrosilation and isomerization reactions. Compound 2c (R3Si = MeCl2Si) was fully characterized via multinuclear NMR spectroscopy and x-ray crystal structure anal. The facile H-transfer rather than Si-transfer to bound COD provides exptl. support for the sequence of insertive steps in the Chalk-Harrod catalytic cycle, at least for Pt-catalyzed hydrosilation.
