All compounds were tested in vitro for their inhibition activity against the human (h) Carbonic Anhydrase (CA; EC 4.2.1.1) I, II, VII, IX and XII isoforms. Among the series, derivatives 1 and 11 showed great enhancement of both inhibition potency and selectivity towards the hCA VII isoform, when compared to the reference SLT drug. The binding mode of 11 within the hCA VII active site was deciphered by means of X-ray crystallog. and revealed the sultam moiety being exposed to the rim of the active site. In vivo experiments on a model of neuropathic pain induced by oxaliplatin clearly showed 11 being an effective pain relieving agent and therefore worth of further exploitation towards the validation of the hCA VII as new target for the management of neuropathies.

Antiviral drug resistance in influenza infections has been a major threat to public health. To develop a broad-spectrum inhibitor of influenza to combat the problem of drug resistance, we previously identified the highly conserved E339…R416 salt bridge of the nucleoprotein trimer as a target and compound 1 as an inhibitor disrupting the salt bridge with an EC50 = 2.7μM against influenza A (A/WSN/1933). We have further modified this compound via a structure-based approach and performed antiviral activity screening to identify compounds 29 and 30 with EC50 values of 110 and 120 nM, resp., and without measurable host cell cytotoxicity. Compared to the clin. used neuraminidase inhibitors, these two compounds showed better activity profiles against drug-resistant influenza A strains, as well as influenza B, and improved survival of influenza-infected mice.

Drug resistant tuberculsosis (TB) is global health crisis that demands novel treatment strategies. Bacterial ATP synthase inhibitors such as bedaquiline and next-generation analogs (such as TBAJ-876) have shown promising efficacy in patient populations and preclin. studies, resp., suggesting that selective targeting of this enzyme presents a validated therapeutic strategy for the treatment of TB. In this work, we report tetrahydronaphthalene amides (THNAs) as a new class of ATP synthase inhibitors that are effective in preventing the growth of Mycobacterium tuberculosis (M.tb) in culture. Design, synthesis and comprehensive structure-activity relationship studies for approx. 80 THNA analogs are described, with a small selection of compounds exhibiting potent (in some cases MIC90 <1 μg/mL) in vitro M.tb growth inhibition taken forward to pharmacokinetic and off-target profiling studies. Ultimately, we show that some of these THNAs possess reduced lipophilic properties, decreased hERG liability, faster mouse/human liver microsomal clearance rates and shorter plasma half-lives compared with bedaquiline, potentially addressing of the main concerns of persistence and phospholipidosis associated with bedaquiline.

We report the synthesis of polymethylene waxes, a surrogate of PE waxes, by a controlled polymerization reaction in water at or near r.t. and under atm. pressure. The monomer, dimethylsulfoxonium methylide, is generated in situ from a salt, trimethylsulfoxonium halide. The carbon sources for the polymerizations are C1 mols., which can be derived from nonpetroleum feedstock. DMSO serves as the C1 carrier and is not consumed. The reaction is initiated and catalyzed by trialkylboranes, compounds that are stable in water. A certain degree of mol. weight control is achieved by adjusting the stoichiometry of “salt” to organoborane. Polymethylene, the simplest hydrocarbon polymer, is a semicrystalline material. The room temperature polymerization produces a linear polymer approx. 100°C below its melting temperature (Tm). The supercooled polymers rapidly crystallize into flat nanoparticles comprised of stacked lamellae.

Those containing CCl3 groups were effective at both high and low loads or only at high and only at low ones. The difference was assumed to be due to different chem. and adsorption activity of the compounds and chem. modification of the friction surface. The effectiveness depended on the concentration of additive (1.5-6 mmole/100 g oil).
