Activity of leading exemplars 19 and 27 in an in vivo model suggestive of therapeutic potential is highlighted herein.

We demonstrate that the cytotoxicity of dansylated polytheonamide mimic is controlled by chem. modification of its N-terminal structure. Dansylated polytheonamide mimic is an ion channel peptide which displays potent cytotoxicity against P388 mouse leukemia cells (IC50 = 12 nM). To modulate its cytotoxicity, three analogs of dansylated polytheonamide mimic, possessing distinct N-terminal structures with different hydrophobicities, were synthesized and their cytotoxicities were evaluated. This focused structure-activity relationship study unveiled that the cytotoxicity of dansylated polytheonamide mimic is enhanced 10-fold by simply changing its N-terminal 5,5-dimethyl-2-oxohexanamide to the more hydrophobic palmitamide. The data obtained here provide new understanding for the functional control of the artificial ion channel peptide.

Newly designed and synthesized diarylethene (DAE) derivatives with aliphatic amine sidearms I and one with two pyrenes II, revealed excellent photo-switching property of central DAE core in MeOH and water. The only exception was bis-pyrene analog, its DAE core very readily photochem. closed, but reversible opening completely hampered by aromatic stacking interaction of pyrene(s) with cyclic DAE. In this process, pyrene fluorescence showed to be a reliable monitoring method, an open form characterized by strong emission at 480 nm (typical for pyrene-aggregate), while closed form emitted weakly at 400 nm (typical for pyrene-DAE quenching). Only open DAE-bis-pyrene form interacted measurably with ds-DNA/RNA by flexible insertion in polynucleotide grooves, while self-stacked closed form did not bind to DNA/RNA. For the same steric reasons, flexible open DAE-bis-pyrene form was bound to at least three different binding sites at bovine serum albumin (BSA), while rigid, self-stacked closed form interacted dominantly with only one BSA site. Preliminary screening of antiproliferative activity against human lung carcinoma cell line A549 revealed that all DAE-derivatives were non-toxic. However, bis-pyrene analog efficiently entered cells and located in the cytoplasm, whereby irradiation by light (315-400 nm) resulted in a strong, photo-induced cytotoxic effect, typical for pyrene-related singlet oxygen species production

The authors describe the efficient synthesis and one-step derivatization of novel, nonfluorescent azo dyes based on the Black Hole Quencher-3 (BHQ-3) scaffold. These dyes were equipped with various reactive and/or bioconjugatable groups (azido, α-iodoacetyl, ketone, terminal alkyne, vicinal diol). The azido derivative is highly reactive in the context of copper-catalyzed azide-alkyne cycloaddition (CuAAC) reactions and allowed easy synthetic access to the first water-soluble (sulfonated derivative) and aldehyde-modified BHQ-3 dyes, the direct preparation of which failed by conventional azo-coupling reactions. The aldehyde- and α-iodoacetyl-containing fluorescence quenchers were readily conjugated to aminooxy- and cysteine-containing peptides by the formation of a stable oxime or thioether linkage, resp. Further fluorescent labeling of the resultant peptide conjugates with red- or far-red-emitting rhodamine or cyanine dyes through sequential and/or one-pot bioconjugations, led to novel Foerster resonance energy transfer (FRET) based probes suitable for the in vivo detection and imaging of urokinase plasminogen activator, a key protease in cancer invasion and metastasis.

The fascinating architecture of these partially-enclosed chiral basket mols. is evident in the solid-state structures which have been determined by single-crystal X-ray crystallog. Moreover, the enantiomers of these chiral resorcinarene crowns have been resolved via diastereomeric resolution, with the absolute configuration of the diastereomers being determined by X-ray crystallog. This work enables further exploration of these enantio-pure chiral baskets as possible chiral resolving agents.
