In this work, a new generation of hydrophobic deep eutectic solvents (DESs) was prepared using eugenol (as hydrogen bond donor) and benzyltriethylammonium bromide, benzyltributylammonium bromide, benzyltriethylammonium chloride and benzyltributylammonium chloride (as hydrogen bond acceptor) in different molar ratios. These DESs were applied to vortex-assisted dispersive liquid-liquid microextraction of Sudan dyes from food samples, followed by high-performance liquid chromatog. determination The influencing parameters, including the type of DES, amount of DES, extraction time, solution pH and salt addition, were investigated and optimized. Under the optimized conditions, a linear range of 2-1000 ng mL-1 with determination coefficients of <0.999 was obtained. Limits of detection and limits of quantification were in the range of 0.5 to 1 ng mL-1 and 2 to 3 ng mL-1, resp. The proposed method was successfully used in the determination of Sudan dyes in chili sauce, chili powder and ketchup, and satisfactory recoveries of between 89.9 and 119.3% were obtained, with relative standard deviations in the range of 0.1-6.8%. The proposed method is simple, green and efficient, and can be applied to determine Sudan dyes in complex matrixes.

Diabetes mellitus is one of the most chronic metabolic disorders. Since past few years, our research group had synthesized and evaluated libraries of heterocyclic compounds against α and β-glucosidase enzymes and found encouraging results. The current study comprises of evaluation of indane-1,3-dione as antidiabetic agents based on our previously reported results obtained from closely related moiety isatin and its derivatives A library of twenty three indane-1,3-dione derivatives (1-23) was synthesized and evaluated for α and β-glucosidase inhibitions. Moreover, in silico docking studies were carried out to investigate the putative binding mode of selected compounds with the target enzyme. The indane-1,3-dione derivatives (1-23) were synthesized by Knoevenagel condensation of different substituted benzaldehydes with indane-1,3-dione under basic condition. The structures of synthetic mols. were deduced by using different spectroscopic techniques, including 1H-, 13C-NMR, EI-MS, and CHN anal. Compounds (1-23) were evaluated for α and β-glucosidase inhibitions by adopting the literature protocols. Off twenty three, eleven compounds displayed good to moderate activity against α- glucosidase enzyme, nonetheless, all compounds exhibited less than 50% inhibition against β- glucosidase enzyme. Compounds 1, 14, and 23 displayed good activity against α-glucosidase enzyme with IC50 values of 2.80 ± 0.11, 0.76 ± 0.01, and 2.17 ± 0.18 μM, resp. The results have shown that these compounds have selectively inhibited the α-glucosidase enzyme. The in silico docking studies also supported the above results and showed different types of interactions of synthetic mols. with the active site of enzyme. The compounds 1, 14, and 23 have shown good inhibition against α-glucosidase and may potentially serve as lead for the development of new therapeutic representatives.

In total, 1936 chems. were evaluated using far-field mass balance human exposure models (USEtox, RAIDAR) and an indicator for indoor and/or consumer use. These predictions were compared to exposures inferred by Bayesian anal. of urine concentrations for 82 chems. reported in the National Health and Nutrition Examination Survey (NHANES). Joint regression of all factors provided a calibrated consensus prediction, the variance of which served as an empirical determination of uncertainty to prioritize absolute exposure potential. Information on use was most predictive; generally, chems. above the limit of detection in NHANES had consumer/indoor use. Coupled with hazard HTS, exposure HTS can assign risk earlier in decision processes. High-priority chems. become targets for further data collection.
