A series of newer benzylideneamino(pyrazinyl)triazole-thiols I [R = 2-HOC6H4, 2,4-di-HOC6H3, 4-FC6H4, 2-HO-naphthyl, etc.] and benzyl(pyrazinyl)sulfanyl-triazolyl-thioureas II [R = Ph, 4-FC6H4, 4-ClC6H4, 2-naphthyl, etc.] a triazole fused with pyrazine moiety of pharmacol. significance was synthesized. All the synthesized compounds were screened for their in vitro antileishmanial and antioxidant activities. Compounds I [R = 4-HOC6H4] (IC50 = 79.0 μM) and II [R = 2,4-di-ClC6H3] (IC50 = 79.0 μM) were shown significant antileishmanial activity when compared with standard sodium stibogluconate (IC50 = 490.0 μM). Compounds I [R = 4-Me2NC6H4] (IC50 = 13.96 μM) and II [R = 3-FC6H4] (IC50 = 13.96 μM) showed significant antioxidant activity. After performing mol. docking study and analyzing overall binding modes it was found that the synthesized compounds had potential to inhibit L. donovani pteridine reductase 1 enzyme.
