In this study, two series of poly(arylene ether sulfone)s (PAES) with hydrophobic blocks made of 4,4′-dihydroxyphenyl (BP) and 2,2-bis(4-hydroxyphenyl) propane (BPA) were synthesized. The copolymers were prepared by a coupling reaction between decafluorobiphenyl (DFBP) end-capped nonsulfonated block (PAES A or B) and sulfonated block (SPAES B). The reactive behavior of DFBP facilitates the low-temperature coupling reaction (below 105°C). The final copolymers were blended at distinct weight percent ratios (0, 20, 30, and 40) with poly(styrene-b-isobutylene-b-styrene) (SIBS) at a sulfonation level of 88% to obtain strong and flexible membranes. Morphol., thermal stability, and conduction properties of the resulting membranes were measured and reported here. Results suggest that phase segregation between hydrophobic and hydrophilic domains occurs below 1 meq. g-1 ion exchange capacity. These membranes have low water absorption and high thermal stability. Incorporating SIBS 88 into the blend added elastomeric behavior enhancing transport properties and addnl. ionic domains, achieving proton conductivities of 0.21 S cm-1 for BPA-based membranes at 50°C (40% SIBS 88). The selectivity of the studied membranes surpasses Nafion 117, making them strong candidates for proton exchange membranes in direct methanol fuel cell applications. Whereas significant racemization was initially observed if the ‘minimalist’ protocol was applied for radiolabeling, it was completely suppressed by the careful adjustment of 18F -preprocessing. The initial biol. study revealed a higher uptake of 2-[ 18F]FPhe in different tumor cells in comparison to that of [ 18F]FET. In contrast to 4-[ 18F]FPhe, which suffered from rapid defluorination in vivo, 2-[ 18F]FPhe demonstrated a sufficient in vivo stability. Conclusively, 2-[ 18F]FPhe is a promising PET probe that is now readily available using Cu-mediated radiofluorination under ‘minimalist’ or ‘low base’ conditions. The simplicity of the translation of the proposed procedures to automated synthesis modules allows a broad biol. evaluation of 2-[ 18F]FPhe. To capture glutaminolysis in tumors, 18F-(2S,4R)4-fluoroglutamine ([18F]FGln) and 18F-fluoroboronoglutamine ([18F]FBQ) have been successfully developed for positron emission tomog. (PET) imaging, but these two mols. lack stability, resulting in undesired yet significant bone uptake. In this study, we found that [18F]FBQ-C2 is a stable Gln PET tracer by adding two more methylene groups to the side chain of [18F]FBQ. [18F]FBQ-C2 was synthesized with a good radiochem. yield of 35% and over 98% radiochem. purity. [18F]FBQ-C2 showed extreme stability in vitro, and no defluorination was observed after 2 h in phosphate buffered saline at 37°C. The competitive inhibition assay results indicated that [18F]FBQ-C2 enters cells via the system ASC and N, similar to natural glutamine, and can be transported by tumor-overexpressed ASCT2. PET imaging and biodistribution results indicated that [18F]FBQ-C2 is stable in vivo with low bone uptake (0.81 ± 0.20% ID/g) and can be cleared rapidly from most tissues. Dynamic scan and pharmacokinetic studies using BGC823-xenograft-bearing mice revealed that [18F]FBQ-C2 accumulates specifically in tumors, with a longer half-life (101.18 ± 6.50 min) in tumor tissues than in other tissues (52.70 ± 12.44 min in muscle). Biodistribution exhibits a high tumor-to-normal tissue ratio (4.8 ± 1.7 for the muscle, 2.5 ± 1.0 for the stomach, 2.2 ± 0.9 for the liver, and 17.8 ± 8.4 for the brain). In conclusion, [18F]FBQ-C2 can be used to perform high-contrast Gln imaging of tumors and can serve as a PET tracer for clin. research. Microsomal metabolic products were identified and quantified by HPLC, chem. assays, and synthesized reference compounds Metabolites identified include products from aromatic ring hydroxylation, side-chain C-hydroxylation (benzyl alcs. and benzaldehydes), N-hydroxylation (hydroxylamines, nitrosotoluenes, azoxy, azo, and hydrazo derivatives), and two new types of microsomal metabolites: secondary amines [i.e. (halogenated) N-(4′-aminobenzyl)-4-toluidines] and an imine [i.e. N-(4′-amino-3′,5′-dichlorobenzylidene)-2,6-dichloro-4-toluidine]. The secondary amine appeared to be a major microsomal metabolite for all three 4-toluidines studied. Quantification of the metabolite patterns demonstrated several influences of the chlorine substituents on metabolism of the 4-toluidine derivatives The total conversion increased significantly in the order: 4-toluidine < 2-chloro-4-toluidine < 2,6-dichloro-4-toluidine, especially because of a marked increase in microsomal side-chain C-hydroxylation with increasing number of ortho chlorine substituents. The rate of N-hydroxylation varied much less. Aromatic ring hydroxylation was observed to a significant extent only for nonchlorinated 4-toluidine. Addnl. data from microsomal binding studies and MO calculations provided insight in possible mechanisms underlying the observed changes in metabolic profiles with increasing number of chlorine substituents at an ortho position with respect to the amine group.
