In this study, solid-phase extraction (SPE) combined with the dispersive liquid-liquid microextraction based on novel hydrophobic deep eutectic solvent (DLLME – DES) has been developed as an ultra-pre-concentration technique for the extraction of aflatoxin M1 (AFM1) in milk-based baby food (MBBF) and infant formula milk (IFM) samples followed by HPLC combined with fluorescence detection (HPLC – FL). In addition, carcinogenic and non-carcinogenic risk assessment was performed by health-related risk factors including liver cancer risk (LCR), margin of exposure (MOE) and target hazard quotient (THQ) were calculated using the mean of AFM1 in different infant food samples. The results of the study showed that the mean of AFM1 was statistically significant different between various brands and types of IFM and MBBF. The results of the study showed that the percentage of pos. samples higher than the allowable limit of AFM1 in 36 samples of domestic infant formula milk (DIFM), 24 samples of imported infant formula milk (IIFM), 36 samples of domestic milk-based baby food (DMBBF) and 18 samples of imported milk-based baby food (IMBBF) were 41.6, 12.5, 66.7 and 33.3%, resp. In addition, estimated values for health risk-related factors including LCR, MOE and THQ indicated that for most infants less than one-year-old were higher than the acceptable levels. Based on the results, it can be concluded that the quality of IFM and MBBF consumed in Iran in terms of AFM1 is poor. Based on the climate data of Kocaeli province in Turkey, an evacuated tube collector is employed to benefit from solar energy to meet the generator heat load of the system. Although at an evaporator temperature of 5 °C, enhanced thermodn. performance is evident with the use of the LiBr +LiCl/H2O system, and a diminished solar collector area is required compared to the system utilizing LiBr/H2O; there is a level of attrition relating to the impact of the former with a single degree rise in evaporator temperature However, this remained at a greater value than for the latter system. The final results pointed out that LiBr+LiCl/H2O has a 48.93% lower circulation ratio, 8.81% higher coefficient of performance (COP) of chiller, 8.88% higher solar COP, 8.96% higher exergy efficiency of chiller, 8.90% higher exergy efficiency of solar-driven system, 8.92% lower solar collector area, and 8.91% lower storage tank volume than LiBr/H2O system in the investigated operating temperature ranges. Surface modification by different quaternary ammonium compounds (QAC) makes nanoclays more compatible with various polymeric matrixes, thereby expanding their potential applications. The growing industrial use of nanoclays could potentially pose a health risk for workers. Here, we assessed how surface modification of nanoclays modulates their pulmonary toxicity. An in vitro screening of the unmodified nanoclay Bentonite (montmorillonite) and four organomodified nanoclays (ONC); coated with various QAC, including benzalkonium chloride (BAC), guided the selection of the materials for the in vivo study. Mice were exposed via a single intratracheal instillation to 18, 54, and 162 μg of unmodified Bentonite or dialkyldimethyl-ammonium-coated ONC (NanofilSE3000), or to 6, 18, and 54 μg of a BAC-coated ONC (Nanofil9), and followed for one, 3, or 28 days. All materials induced dose- and time-dependent responses in the exposed mice. However, all doses of Bentonite induced larger, but reversible, inflammation (BAL neutrophils) and acute phase response (Saa3 gene expression in lung) than the two ONC. Similarly, highest levels of DNA strand breaks were found in BAL cells of mice exposed to Bentonite 1 day post-exposure. A significant increase of DNA strand breaks was detected also for NanofilSE3000, 3 days post-exposure. The sphingomyelin synthase 2 (SMS2) is a potential target for pharmacol. intervention in atherosclerosis. However, so far, few selective SMS2 inhibitors and their pharmacol. activities were reported. The selectivity ratio of Ly93 was more than 1400-fold for purified SMS2 over SMS1. The in vitro studies indicated that Ly93 not only dose-dependently diminished apoB secretion from Huh7 cells, but also significantly reduced the SMS activity and increased cholesterol efflux from macrophages. Meanwhile, Ly93 inhibited the secretion of LPS-mediated pro-inflammatory cytokine and chemokine in macrophages. The pharmacokinetic profiles of Ly93 performed on C57BL/6J mice demonstrated that Ly93 was orally efficacious. As a potent selective SMS2 inhibitor, Ly93 significantly decreased the plasma SM levels of C57BL/6J mice. Furthermore, Ly93 was capable of dose-dependently attenuating the atherosclerotic lesions in the root and the entire aorta as well as macrophage content in lesions, in apolipoprotein E gene knockout mice treated with Ly93. Indole is an important organic synthesis intermediate derived from the wash oil fraction in coal tar. Deep eutectic solvents (DESs) have been widely used in the separation field due to their unique properties such as easy preparation, low cost, green and high extraction efficiency, and reusability. In this study, five DESs, including tetramethylammonium chloride/ethylene glycol (TMAC/EG), triethylmethylammonium chloride/ethylene glycol (TEMAC/EG), tetraethylammonium chloride/ ethylene glycol (TEAC/EG), tetrapropylammonium chloride/ethylene glycol (TPAC/EG), tetrabutylammonium chloride /ethylene glycol (TBAC/EG) were prepared and used to extract indole from model wash oil. The results showed that TPAC/EG exhibited the best extraction ability under the same conditions. Then, the main exptl. parameters such as mass ratio of DES to model wash oil, extraction temperature, extraction time, standing time and initial concentration of indole, were investigated when using TPAC/EG as extractant. In optimal conditions (25°C, 5 min, mass ratio 1:8), the extraction efficiency, selectivity and distribution coefficient for indole were 90.51%, 79.21% and 61.12, resp. Moreover, removing neutral oil with petroleum ether, the purity of indole further reached 96.63%, which was much higher than that in the previous studies reported. In addition, accessible and environmentally friendly water instead of di-Et ether was used to regenerate the DES. Reused four times, the TPAC/EG extraction ability for indole almost did not change. Finally, the mechanism of extraction process was investigated by FT-IR anal. This work provides a new route for extraction and recovery indole from wash oil. Treatment of two molar equivalents of 1,8-diaminonaphthalene-2-thione with one molar equivalent of the metal’s(II) salt (Pd, Pt, Hg, Cd, Zn) afforded both neutral thione complexes and thionate derived complexes. The prepared ligand and its complexes (18) were characterized via different spectroscopic technics including: FTIR, 1H, 13C-NMR, and mass spectroscopy. The spectroscopic data anal. suggested that these compounds adopt different structural geometries. For instance, Pd(II) and Pt(II) complexes show square planar geometry. Whereas, thione complexes of Hg(II), Zn(II), and Cd(II) showed probable tetrahedral structure. However, thionate derived complexes of Hg, and Ph-Hg are expected to be linear. The thionato-Cd(II) complex show bidentate chelating mode through its N and S donor atoms. Furthermore, the newly prepared complexes exhibit nano-structural characteristics, which were measured by SEM and x-ray diffraction of the powder. SEM data showed that complexes from (3, 5) and (8) gave irregular nano-structure, nano-rod structure, and nano-sphere structural pattern, resp. However, Complexes (1) and (2) were tested against MCF-7 (breast) and LoVo (colon) cancer cells using MTT assay. The palladium complex showed a promising antiproliferative activity against colon (LoVo) and breast (MCF-7) cancer cells with IC50 values 21.13 and 22.25μM, resp. In addition the antibacterial activity was study against two different pathogenic bacteria, and the Pd(II) complex displayed highest activity compared with other complexes. The plasma membrane (NCX) and mitochondrial (NCLX) Na+/Ca2+ exchangers are structurally related proteins, although they operate under strictly different ionic conditions and membrane potentials. In contrast with NCX, NCLX can transport either Li+ or Na+ in exchange for Ca2+. Whereas the crystal structure of the archaeal NCX (NCX_Mj) describes the binding sites for alternative binding of 3Na+ or 1Ca2+, these features remain elusive for NCLX due to the lack of structural information. To elucidate the ion-binding features of mitochondrial NCLX, we analyzed here the Li+-transporting NCLX_Mj mutant, produced by replacing the ion-coordinating residues in the archaeal NCX (NCX_Mj) to match the ion-coordinating residues of human NCLX. The NCLX_Mj-mediated Na+/Ca2+ or Lia+/Ca2+ exchange rates are insensitive to varying voltage, consistent with an electroneutral ion exchange. Mol. dynamics (MD) simulations revealed that NCLX_Mj contains two novel Li+ binding sites with four ion-coordinating residues, derived from the three Na+ binding sites of NCX_Mj. The ion-coordination modes, observed in the MD simulations, were further supported by two-dimensional IR (2D IR) spectroscopy and by testing the mutational effects on the ion fluxes. Collectively, our results revealed a structural basis for Li+ binding and electroneutral transport (2Na+/Li+:1Ca2+) by NCLX_Mj, meaning that the NCLX-mediated electroneutral transport may predefine mitochondrial Ca2+ and Na+signaling to modulate cellular functions.
