Photoactivatable antimicrobial compounds and methods for the use thereof in the treatment of infections are provided. Photosensitizers, drugs, and targeting moieties are conjugated using linkers which comprise an enzyme cleavage site for an enzyme of a pathogen. The enzyme cleavage site comprises a cephalosporin, penicillin, penem, etc. The targeting moiety may be directed to a pathogen or a host cell infected with a pathogen. Photosensitizers are present in an amount sufficient to quench photoactivation of the photosensitizers. An example is provided of the preparation of a cephalosporanic acid-photosensitizer prodrug which is cleaved in the presence of β-lactamase from Enterobacter cloacae.
