The current report represents a transition-metal-free synthesis of oxazoline and isoxazoline derivatives by a tandem electro-oxidative chalcogenation-cyclization process. Both C-Se and C-S bond-forming protocols were developed without using any external oxidant and the reaction was performed at room temperature, open to the air. Using this methodology, 29 substituted oxazoline and 16 substituted isoxazoline derivatives were synthesized with up to 91% isolated yield.

A simple, efficient, and metal-free methodology for the preparation of 2-mercaptobenzothiazole and derivatives in excellent yields via microwave-assisted pathway is reported. Our condition provides a convenient protocol for the synthesis of a diverse collection of 2-mercaptobenzothiazoles and 6-(4-substituted-1H-1,2,3-triazol-1-yl)-2-mercaptobenzothiazoles with a very simple purification process. This report provides an alternative protocol for fast access to the wide range of compounds for sequence synthesis and biological studies.

An unusual phenomenon occurred in new display cases at the Rijksmuseum four months after their installation in April 2013. White deposits were visible on glass windows, silicone door gaskets, black structural adhesive seals, and on works of art. The works of art most affected by these deposits were bronze sculptures, wooden and waxed objects, tempera, and oil paintings. In this study the deposits were chemically characterised using (pyrolysis-)gas chromatography-mass spectrometry ((Py-)GC-MS), mu-Raman spectroscopy (mu-Raman), ion chromatography (IC), and scanning electron microscopy combined with energy dispersive X-ray spectroscopy (SEM-EDX). Five chemically different crystalline deposits were found. They were identified as organic salts of the base 2,2,6,6-tetramethyl-4-piperidinol (TMP-ol), a secondary amine, and five different carboxylic acids. It was found that TMP-ol, which is part of the UV-light stabiliser Tinuvin-770, emitted from the structural adhesive Terostat-9220. Terostat-9220 was used in large quantities in the display cases to adhere glass windows to metal parts. The carboxylic acids derived from both construction materials used to build the cases and from conservation materials present on the exhibited works of art. The carboxylic acids involved were 2,4-dichlorobenzoic acid, formic acid, methacrylic acid, palmitic acid, and an unknown carboxylic acid, respectively emitted from peroxide-cured silicone gaskets, panels of medium-density fibreboard (MDF), UV-adhesive, beeswax containing products, and from an unknown acidic conservation product or binding medium. The identification of the crystalline deposits was supported by their syntheses in the laboratory. Since 2013, similar deposits have been observed in a number of museum collections worldwide. A treatment for preventing further growth of the deposits was developed and applied in the Rijksmuseum showcases.

Much of the worlds groundwater supply has been contaminated by aromatic hydrocarbons originating from anthropogenic sources. To study the occurrence and distribution characteristics of aromatic hydrocarbons in groundwater, 24 aromatic hydrocarbon compounds were selected: Five BTEX compounds (benzene, toluene, ethylbenzene, o-xylene, m-xylene, and p-xylene), 10 alkyl-substituted benzene, and 9 halogenated aromatics. These aromatic hydrocarbons were then analyzed from 355 samples collected from across China. Results indicated that aromatic hydrocarbons were detected in 59 out of 355 samples. Of the selected aromatic hydrocarbons, BTEX compounds were detected with high frequency and at low concentrations; comparatively, halogenated aromatics were detected with low frequency and at high concentrations. The aromatic hydrocarbon characteristics found in both karst and pore groundwater samples were then determined using their respective hydrogeological conditions and corresponding human activities. In karst groundwater, BTEX compounds were the most frequently detected aromatic hydrocarbon. The high detection frequencies of aromatic hydrocarbons were caused by their rapid migration, owing to the developed conduit system in the sampled karst area. The low concentrations of aromatic hydrocarbons in karst groundwater samples were caused by low-intensity human activity along with special hydrogeological conditions with higher redox potential and the unique compositions of aromatic hydrocarbons. Alkyl-substituted aromatics and halogenated aromatics were detected at higher concentrations in pore groundwater, owing to high-intensity human activity. Aromatic hydrocarbon pollution was gradually decreased along piedmont-alluvial plain-coast line, owing to a decrease in aquifer vulnerability. These were positively correlated with the size of the aquifer’s particles. Samples with a high accumulative concentration of these aromatic hydrocarbons tended to occur in pore groundwater with a high concentration of either SO42- or Cl-.

Background:Licensed therapies for nonalcoholic fatty liver disease (NAFLD) do not yet exist, but clinical trials are testing treatment options. Inclusion criteria often require liver biopsy showing fibrosis (F2/3) or cirrhosis (F4) and nonalcoholic steatohepatitis (NASH). However, histological criteria pose a serious obstacle for recruitment. Aims:Characterize the relevance of liver biopsies in the selection of patients with NAFLD. Methods:Patients between 2013 and 2018 with the ICD-10 code K76.0 were analyzed. Fibrosis was defined by the NASH clinical research network (CRN) fibrosis staging system, NASH by a NAFLD activity score (NAS) >= 4. Predictive factors were determined by logistic regression. Results:Liver biopsy was performed in 87/638 (13.6%) patients (49% female, age 52.5 +/- 14.0, BMI 30.4 +/- 5.9 kg/m(2)). Fibrosis stage F0/F1/F2/F3/F4 was observed inN = 7/47/7/17/9, an NAS >= 4 inN = 27. Fibrosis stage F2/F3 and F4 along with NAS >= 4 was found in 1.7% and 0.5% of cases. Liver stiffness measurement, LSM (OR 2.3 per doubling of value; CI 1.3-4.4,p = .005) and FIB-4 (OR 2.3 per doubling of value; CI 1.2-4.4,p = .012) were significant predictors for fibrosis >= F2. Predictive factors for NASH were not identified. Conclusion:The biopsy rate in NAFLD patients is low and fibrosis >= F2 along with NAS >= 4 only present in a few cases. Transient elastography and FIB-4 are useful to select patients at risk for fibrosis for liver biopsy.

Toward the search of new antibacterial agents to control methicillin-resistant Staphylococcus aureus (MRSA), a class of new norfloxacin-1,3,4-oxadiazole hybrids were designed and synthesized. Antibacterial activities against drug-sensitive bacteria S. aureus and clinical drug resistant isolates of MRSA were evaluated. Compound 5k exhibited excellent antibacterial activities against S. aureus (MIC: 2 mu g/mL) and MRSA1-3 (MIC: 0.25-1 mu g/mL). The time-kill kinetics demonstrated that compound 5k had an advantage over commonly used antibiotics vancomycin in killing S. aureus and MRSA. Moreover, compound 5k could inhibit the bacteria and destroy their membranes in a short time, and showed very low cytotoxicity to NRK-52E cells. Some interesting structure-activity relationships (SARs) were also discussed. These results indicated that these norfloxacin-1,3,4-oxadiazole hybrids could be further developed into new antibacterial agents against MRSA.

This paper demonstrates a selective, mild approach to Ullmann amination of aryl halides to synthesize N-alkylated derivatives of epsilon-caprolactam. The synthetic route involves an in-situ ring-opening of 1,8-diazabicyclo[5.4.0]undec-8-ene (DBU) followed by concurrent arylation with aryl halides in the presence of copper iodide as a catalyst under ligand-free conditions. This method provides a new entry to a wide variety of epsilon-caprolactam derivatives in good to excellent yields in a single synthetic sequence. Similarly, other bicyclic amidines such as 1, 5-diazabicyclo-[4.3.0]non-5-ene (DBN), and 1,5,7 triazabicyclo[4.4.0] dec-5-ene (TBD) also showed good to very high reactivity. Multitargeted hybrids of N-benzylpiperidine and substituted 5-phenyl-1,3,4-oxadiazoles were designed, synthesized, and evaluated against Alzheimer’s disease (AD). Tested compounds exhibited moderate to excellent inhibition against human acetylcholinesterase (hAChE), butyrylcholinesterase (hBChE), and beta-secretase-1 (hBACE-1). The potential leads 6g and 10f exhibited balanced inhibitory profiles against all the targets, with a substantial displacement of propidium iodide from the peripheral anionic site of hAChE. Hybrids 6g and 10f also elicited favorable permeation across the blood -brain barrier and were devoid of neurotoxic liability toward SH-SYSY neuroblastoma cells. Both leads remarkably disassembled A beta aggregation in thioflavin T-based selfand AChE-induced experiments. Compounds 6g and 10f ameliorated scopolamine gamma-induced cognitive dysfunctions in the Ymaze test. The ex vivo studies of rat brain homogenates established the reduced AChE levels and antioxidant activity of both compounds. Compound 6g also elicited noteworthy improvement in A beta-induced cognitive dysfunctions in the Morris water maze test with downregulation in the expression of A beta and BACE-1 proteins corroborated by Western blot and immunohistochemical analysis. The pharmacokinetic study showed excellent oral absorption characteristics of compound 6g. Environmental transformation products of pesticides (ETPPs) have a great deal of ecological impact owing to their ability to cause toxicity to the aquatic organisms, which can then be translated to the humans. The limited experimental data on biochemical and toxic effects of ETPPs, the high test costs together with regulatory limitations and the international push to reduce animal testing encourage greater dependence on predictive in silico techniques like quantitative structure-activity relationship (QSAR) models. The aim of the present work was to explore the key structural features, which regulate the toxicity towards fishes, for 85 ETPPs using a partial least squares (PLS) regression based chemometric model developed according to Organisation for Economic Co-operation and Development (OECD) guidelines. The model was extensively validated using both internal and external validation metrics, and the results so obtained justify the reliability and usefulness of the developed model (Q(2) = 0.648, R-pred(2) or Q(F1)(2) = 0.734 and Q(F2)(2) = 0.733). From the developed model, we can conclude that lipophilicity, polarity, presence of branching and the functional form of 0-atom in the transformed structures of pesticides are the important features that are to be considered during ecotoxicity assessment of ETPPs. The information obtained from the descriptors of the developed model could be utilized in the future for assessing ETPPs with the benefit of providing an early warning of their potentially detrimental effect on fishes for regulatory purposes. We report the catalytic reduction of both active esters and amides by selective C(acyl)-X (X=O, N) cleavage to access aldehyde functionality via a palladium-catalyzed strategy. Reactions are promoted by hydrosilanes as reducing reagents with good to excellent yields and with excellent chemoselectivity for C(acyl)-N and C(acyl)-O bond cleavage. Carboxylic acid C(acyl)-O bonds are activated by 2-chloro-4,6-dimethoxy-1,3,5-triazine (CDMT) to form triazine ester intermediates, which further react with hydrosilanes to yield aldehydes in one-pot two-step procedures. We demonstrate that C(acyl)-O cleavage/formylation offers higher yields and broader substrate scopes compared with C(acyl)-N cleavage under the same reaction conditions.
