The A-type Aurora kinase is upregulated in many human cancers, and it stabilizes MYC-family oncoproteins, which have long been considered an undruggable target. Here, we describe the design and synthesis of a series of pyrimidine-based derivatives able to inhibit Aurora A kinase activity and reduce levels of cMYC and MYCN. Through structure-based drug design of a small molecule that induces the DFG-out conformation of Aurora A kinase, lead compound 13 was identified, which potently (IC50 < 200 nM) inhibited the proliferation of high-MYC expressing small-cell lung cancer (SCLC) cell lines. Pharmacokinetic optimization of 13 by prodrug strategies resulted in orally bioavailable 25, which demonstrated an 8-fold higher oral AUC (F = 62.3%). Pharmacodynamic studies of 25 showed it to effectively reduce cMYC protein levels, leading to >80% tumor regression of NCI-H446 SCLC xenograft tumors in mice. These results support the potential of 25 for the treatment of MYC-amplified cancers including SCLC.

The reaction proceeds through base-catalyzed benzyl C-H deprotonation and cyclization to afford 2-aryl-2H-indazoles in good yields. A concurrent tandem catalytic (CTC) methodology has been developed for the amidation of aryl chlorides where the aryl chloride is first converted to an aryl iodide via halogen exchange and the aryl iodide is subsequently transformed into the aryl amide. A variety of aryl chlorides were converted to aryl amides in up to 85% isolated yield using 20 mol % CuI, 60 mol % N,N’-cyclohexane-1,2-diamine, 2.2 equiv of K2CO3, and 1.05-1.5 equiv of amide in acetonitrile at 200 degrees C after 0.75-1 h. The same copper/ligand system served as multifunctional catalyst for both steps of the concurrent catalytic process with iodide present in substoichiometric amounts. Mechanistic studies were consistent with CTC amidation occurring via a nonradical mechanism. Kinetic modeling was conducted to investigate the effect of competitive direct amidation of an aryl chloride or aryl bromide on the formation of product over time during a CTC amidation reaction.

Type I Interferon (IFN) signaling plays an important role in the immune defense system against virus infection and in the innate immune response, thus IFNs are widely used as anti-viral agents and treatment for immune disorder or cancer. However, there is a growing demand for novel small-molecule IFN inducer due to tolerance, toxicity, or short duration of action following direct administration of IFNs. In this study, we assessed arylpiperazine (ARP) as a new core skeleton of IFN inducer. To investigate structure-activity relationship, we designed and synthesized a series of ARP analogues and evaluated the ability to stimulate IFN response in THP-1 human monocyte cells. Compound 5i was identified as a potent type I IFN inducer as it significantly increased cytokine secretion and increased expression of various IFN-stimulating genes which are representative biomarkers of type I IFN pathway. Our results suggested a beneficial therapeutic potential of 5i as an anti-viral agent.

Graphene oxide (GO)-catalyzed selective synthesis of amides via C-Sp3-H activation of methylarenes and consequent C-N bond formation with anilines under aqueous medium has been described. Oxygen functionality allied with GO surface played a dual role both as acid catalyst and oxidizing agent to some extent. However, GO has a copious effect on the reaction, shown by a high TOF value with TBHP as co-oxidant. The decisive role of carboxylic acid functional groups on GO nanosheets in this metal-free strategy has been confirmed and was monitored by various analytic techniques viz. Fourier transform-infrared, UV-Vis, Raman and XPS. A plausible mechanism was proposed by control experiments and by the isolation of the intermediate. Over-oxidation of methylarenes was not detected, and high recyclability of the carbocatalyst with its heterogeneous behavior facilitated the isolation and purification of the desired products. We have further explored the utility of this process for the chemoselective synthesis of benzimidazoles.

In the present study, carbon black (CB), multi walled carbon nanotubes (MWCNTs), and granular activated carbon (GAC) were employed to support palladium (Pd) catalyst. The prepared Pd/CB, Pd/MWCNTs and Pd/GAC moveable catalysts were aimed to address the common issues (easy loss of catalyst and poor long-term performance) of normal fixed catalysts. The results of various characterizations (e.g., TEM, XRD, and XPS) clearly show that the Pd nanopartides were successfully loaded onto the carbon-supports, especially CB and MWCNTs. And more importantly, the morphologies, Pd distribution, and chemical structure of these moveable catalysts were almost not changed after 3 h reaction. The moveable Pd/CB, Pd/MWCNTs, and Pd/GAC catalysts had good reactivity for 2,4-dichlorobenzoic acid (2,4-DCBA) dechlorination, and Pd/CB exhibited the best performance. The Pd/CB also shows the best adsorption capacity of 2,4-DCBA and dechlorination product (benzoic acid, BA), and the adsorption of BA was significantly inhibited in the presence of current due to the repulsion between the both negatively charged compounds and adsorbents. The removal of 2,4-DCBA and the generation rate of BA was improved with a pre-adsorption process, which was a promising strategy with higher dechlorination rate and shortened electrolysis time. Moreover, the loss of Pd catalyst was negligible during the 10 consecutive cycles experiment, and the improved longevity could be expected. These results suggested the good reactivity, stability, and reusability of moveable Pd/CB catalyst.

Graphitic carbon nitride materials have attracted significant interest in recent years and found applications in diverse light-to-energy conversions such as artificial photosynthesis, CO2 reduction, or degradation of organic pollutants. However, their utilization in synthetic photocatalysis, especially in the direct functionalization of C(sp(3))-H bonds, remains underexplored. Herein, we report mesoporous graphitic carbon nitride (mpg-CN as a heterogeneous organic semiconductor photocatalyst for direct arylation of C(sp(3))-H bonds in combination with nickel catalysis. Our protocol has a broad synthetic scope (>70 examples including late-stage functionalization of drugs and agrochemicals), is operationally simple, and shows high chemo- and regioselectivities. Facile separation and recycling of the mpg-CN catalyst in combination with its low preparation cost, innate photochemical stability, and low toxicity are beneficial features overcoming typical shortcomings of homogeneous photocatalysis. Detailed mechanistic investigations and kinetic studies indicate that an unprecedented energy-transfer process (EnT) from the organic semiconductor to the nickel complex is operating.

In this study, a type of thiazolium-labeled 1,3,4-oxadiazole thioether bridged by diverse alkyl chain lengths was constructed. The antimicrobial activity of the fabricated thioether toward plant pathogenic bacteria and fungi was then screened. Antibacterial evaluation indicated that title compounds possess specific characteristics that enable them to severely attack three phytopathogens, namely, Xanthomonas oryzae pv. oryzae, Ralstonia solanacearum, and Xanthomonas axonopodis pv. citri with minimal EC50 values of 0.10, 3.27, and 3.50 mu g/mL, respectively. Three-dimensional quantitative structure-activity relationship models were established to direct the following excogitation for exploring higher active drugs. The in vivo study against plant bacterial diseases further identified the prospective application of title compounds as alternative antibacterial agents. The proteomic technique, scanning electron microscopy patterns, and fluorescence spectrometry were exploited to investigate the antibacterial mechanism. Additionally, some target compounds performed superior inhibitory actions against three tested fungal strains. In view of their simple molecular architecture and highly efficient bioactivity, these substrates could be further explored as promising surrogates for fighting against plant microbial infections.

A series of dimetallic palladium(II)-NHC complexes comprised of 1,4-naphthalenyl or 9,10-anthracenyl spacer sandwiched between two imidazole rings was successfully synthesized. These complexes were characterized by H-1 and C-13{H-1} NMR spectroscopy and elemental analysis. The structures of two dimetallic palladium complexes and a related mononuclear palladium complex to be used for comparative studies were further characterized by X-ray diffraction. The dimetallic palladium complex with the 9,10-anthracenyl linker was very efficient in catalyzing direct C-H arylation reactions of heteroaromatic compounds (imidazoles, imidazo[1,2-a]pyridine, and thioazole) with a broad range of aryl chlorides, employing a mild monopalladium loading of 1.5 mol%. It allows for the effective use of aryl chlorides to prepare arylated heterocycles, previously only accessible with the more reactive bromide counterparts. Importantly, the catalytic activity of the dimetallic precatalyst was found to be higher than that of an analogous mononuclear complex.

We compare the local triage tool and internationally validated tools among under-fives presenting to an urban emergency department in Tanzania.MethodologyProspective descriptive study of consecutive under-fives seen at Muhimbili National Hospital (MNH), ED between November 2017 to April 2018. Patients were triaged according to Local Triage System (LTS), and the information collected were used to assign acuities in the other triage scales: Canadian Triage and Acuity Scale (CTAS), Australasian Triage Scale (ATS), Manchester Triage Scale (MTS) and South African Triage Scale (SATS). Patients were then followed up to determine disposition and 24h outcome. Sensitivity, specificity, positive and negative predictive values for admission and mortality were then calculated.ResultsA total of 384 paediatric patients were enrolled, their median age was 17months (IQR 7-36months). Using LTS, 67(17.4%) patients were triaged in level one, 291(75.8%) level 2 and 26 (6.8%) in level 3 categories. Overall admission rate was 59.6% and at 24h there were five deaths (1.3%). Using Level 1 in LTS, and Levels 1 and 2 in other systems, sensitivity and specificity for admission for all triage scales ranged between 27.1-28.4% and 95.4-98% respectively, (PPV 90.3-95.3%, NPV 47.1-47.4%). Sensitivity for mortality was 80% for LTS, and 100% for the other scales, while specificity was low, yielding a PPV for all scales between 6.9 and 8%.ConclusionAll triage scales showed poor ability to predict need for admission, however all triage scales except LTS predicted mortality. The test characteristics for the other scales were similar. Future studies should focus on determining the reliability and validity of each of these triage tools in our setting.
