An efficient, metal-free C-H halogenated method for the synthesis of beta-halogenated carboxamides using non-corrosive NXS (X=Cl, Br) as the halogenated source and ammonium sulfocyanide (NH4SCN) as the additive was proposed and accomplished. Various substituted 2-oxazolines efficiently afforded the corresponding beta-halogenated carboxamides via a ring-opening reaction in good yields. The characteristic features of this reaction include readily available materials, mild reaction conditions and functional groups tolerance.

Alternatives to the use of pressurized hydrogen (H-2) in chemical syntheses are of growing interest and importance in developing safer protocols. Ammonia borane (AB), with its high hydrogen content and ability to release H-2 under mild conditions, is one potential solution. However, optimization of AB dehydrogenation and further the use of AB as a H-2 source to facilitate tandem one-pot reactions with both high activity and stability is an ongoing research challenge. In this work, monodisperse AuPd nanoparticle (NP) catalysts were synthesized and systematically studied for AB dehydrogenation, and Au-rich NPs were found to be the most active composition for AB methanolysis with a turnover frequency (TOF) of 160 min(-1) surpassing most Pd-based NP catalysts to date. More importantly, Au-rich NPs were also active in the one-pot tandem reduction of a series of nitro-groups and the hydrodehalogenation of mono- and polyhalogenated persistent chemical pollutants under mild reaction conditions using only a 2:1 molar ratio of AB to substrate. The AuPd catalyst was stable after ten reaction runs for AB methanolysis and five reaction runs for tandem one-pot reactions without a loss of activity or change in NP structure and morphology.

In the presence of [p-cymene)RuCl2](2), (E)-configured alkenyl bromides couple with aromatic carboxylates to form orthovinylbenzoic acids. An efficient and operationally simple protocol has been demonstrated for the synthesis of 1,3,5,7-tetrasubstituted pyrimido[4,5-d]pyrimidines via TBHP-mediated direct oxidative coupling of N-uracil amidines and methylarenes under metal-free conditions. Due to the inherent stability of methylarenes compared to aldehydes, the presented synthetic protocol is adaptable to a broad substrate scope, is operationally simple, has no need for stringent protection in the whole preparation process, and has the potential to prepare valuable products that are currently inaccessible or challenging to prepare using conventional methods. Providing new methods for the selective functionalization of small molecules is highly desirable, because installing molecular diversity in a desired position, for example, allows one to modulate bioactive molecules. This work reports a method for the selective functionalization of anilines using hexafluoroisopropanol (HFIP) as a solvent to promote an acid-catalyzed hydroarylation of olefins. Mechanistic experiments revealed that HFIP both protonates the alkene and selectively enables anilines toward the electrophilic aromatic substitution. This powerful strategy has been applied to the functionalization of the anti-inflammatory mefenamic acid with chemocontrol and regiocontrol.

Retinoic acid receptor-related orphan receptor gamma t (ROR gamma t) is a nuclear receptor associated with the pathogenesis of autoimmune diseases. Allosteric inhibition of ROR gamma t is conceptually new, unique for this specific nuclear receptor, and offers advantages over traditional orthosteric inhibition. Here, we report a highly efficient in silico-guided approach that led to the discovery of novel allosteric ROR gamma t inverse agonists with a distinct isoxazole chemotype. The the most potent compound, 25 (FM26), displayed submicromolar inhibition in a coactivator recruitment assay and effectively reduced IL-17a mRNA production in EM cells, a marker of ROR gamma t activity. The projected allosteric mode of action of 25 was confirmed by biochemical experiments and cocrystallization with the ROR gamma t ligand binding domain. The isoxazole compounds have promising pharmacokinetic properties comparable to other allosteric ligands but with a more diverse chemotype. The efficient ligand-based design approach adopted demonstrates its versatility in generating chemical diversity for allosteric targeting of ROR gamma t.

The present study reports on the degradation of chlorinated benzoic acids (CBAs), commonly present in the environment as pollutants, by a hydrodehalogenation reaction utilizing the Raney Al-Ni alloy (50:50 wt% Al:Ni). The hydrodehalogenation reaction using the Raney Al-Ni alloy has already been proven as an efficient tool for fast and efficient degradation of halogenated persistent organic pollutants (POPs). Herein, the nano-structured Raney Al-Ni alloy was prepared by an alternative mechano-thermal approach starting from pure elements in a form of powders. The prepared alloy was characterized by X-ray diffractometry, scanning electron microscopy, particle size distribution, and active surface area analyses. The properties of the material were compared with a commercial sample of the same alloy prepared by the atomization process. The activity of the synthesized alloy was evaluated as removal efficiency and a rate of dehalogenation of three different CBAs – 2-chlorobenzoic acid, 2,6-dichlorobenzoic acid, and 2,3,6-trichlorobenzoic acid (trysben); used in the past as an herbicide. Dehalogenation of all three tested CBAs yielded benzoic acid as the only product and followed the first-order reaction kinetics. Compared to the commercially available alloy, enhanced kinetics of CBAs removal was achieved, owing to the solid-state properties of the mechano-thermally prepared alloy.

Searching for CNS active cyclic amines derivatives containing heterocyclic xanthone core we designed and synthesized a set of fourteen novel 2- or 4-methylxanthone substituted by alkyl- or aryl-piperazine moieties. The compounds were evaluated in vivo for their potential antidepressant-like activity (in the forced swim test) and anxiolytic-like activity (four-plate test) and their inhibitory effect against rat 5-HT2 receptor was checked. The pharmacokinetic analysis of active compounds done by a non-compartmental approach have shown a rapid absorption of all studied molecules from intraperitoneal cavity and good penetration the blood-brain barrier after i.p. administration with brain to plasma ratios varied from 2.8 to 31.6. Genotoxicity and biotransformation of active compounds were studied. Compound 19 interactions with major classes of GPCRs, uptake systems and ion channels were tested and results indicated that it binds to 5-HT2A, 5-HT2B receptors and sodium channels.

Electrocatalytic hydrodechlorination (ECH) has been proposed as a potential technology for the effective degradation of organochlorine contaminants. In this study, a nanoscale Pd/TiC catalyst with low resistance was dip-coated onto Ti, aiming to fabricate a stable Pd/TiC/Ti electrode with enhanced ECH reactivity for efficient dechlorination. Electrochemical techniques indicated that the Pd/TiC/Ti electrode had a larger electrochemically active surface area and more rapid interfacial charge transfer than the conventional Pd/C/Ti electrode. In a batch experiment, the dechlorination rate for ECH of 2,4-dichlorobenzoic acid (2,4-DCBA, a model pollutant) by Pd/TiC/Ti was 1.3-14.5 times higher than those by Pd/C/Ti, Pd/MWCNTs/Ti, and Pd/MoS2/Ti, respectively. Scavenger experiments, ESR spin-trapping spectroscopy, and cyclic voltammetry clarified the intensified atomic H* generation by Pd/TiC/Ti. DFT calculations revealed that Pd/TiC/Ti had stronger binding ability with atomic H* (E-ads: -2.89 eV vs. -2.44 eV) and 2,4-DCBA (E-ads: -3.62 eV vs. -2.15 eV), respectively, when compared with Pd/C/Ti. As a result, enhanced generation of atomic H*, together with strengthened adsorption of atomic H* and 2,4-DCBA, would contribute to a faster ECH reaction. The Pd/TiC/Ti electrode retained its reactivity after 10 successive batch experiments (80 h), and showed high tolerance to the constituents in actual water matrices including lake water and river water. This study proposed a promising ECH electrode for the treatment of chlorinated organics in water matrices.
