A simple, efficient, and metal-free methodology for the preparation of 2-mercaptobenzothiazole and derivatives in excellent yields via microwave-assisted pathway is reported. Our condition provides a convenient protocol for the synthesis of a diverse collection of 2-mercaptobenzothiazoles and 6-(4-substituted-1H-1,2,3-triazol-1-yl)-2-mercaptobenzothiazoles with a very simple purification process. This report provides an alternative protocol for fast access to the wide range of compounds for sequence synthesis and biological studies.

Although 3-azoindoles have recently emerged as an appealing family of photoswitch molecules, the synthesis of such compounds has been poorly covered in the literature. Herein a high-yielding and operationally simple protocol is reported allowing the synthesis of 3-azoindoles, featuring important steric hindrance around the azo motif. Remarkably, this C-H coupling is characterized by excellent atom economy and occurs under metal-free conditions, at room temperature, and within few minutes, delivering the expected products in excellent yields (quantitatively in most of the cases). Accordingly, a library of new molecules, with potential applications as photochromic compounds, is prepared.

Oxathiapiprolin is one of the best active fungicides discovered for oomycetes control. To develop a fungicide candidate with a broad spectrum of activity, 22 new piperidinylthiazole derivatives were designed and synthesized. Compound 51 showed the best activity against Pseudoperonospora cubensis (Berk. et Curt.) Rostov and Phytophthora infestans in vivo with 100% and 80% of inhibition, respectively, at 1 mg/L, and 72.87% of field efficacy against P. cubensis at 1 g ai/667 m(2) validated these results. Compound 5i exhibited a broad spectrum of excellent activity against Sclerotinia sclerotiorum with EC50 = 0.30 mg/ L (>10 times more active than oxathiapiprolin and azoxystrobin in vitro), good activity against Botrytis cinerea, Cercospora arachidicola, and Gibberella zeae with EC50 of 14.54, 5.57, and 14.03 mg/L in vitro and against P. cubensis and P. infestans with 60% and 30% inhibition rates, respectively, at 1 mg/L in vivo. Mode of action studies by RNA sequencing analysis discovered oxysterol-binding protein (OSBP), chitin synthase (CHS1), and (1,3)-beta-glucan synthase (FKS2) as the potent target of 5i against S. sclerotiorum. Quenching studies validated that OSBP was the same target of both 5i and oxathiapiprolin; it was quenched by both of them. Our studies discovered isothiazole-containing piperidinylthiazole as an OSBP target-based novel lead for fungicide development.

In this paper, synthesized Fe3O4 nanoparticles applied as catalyst in photodegradation of Direct Red 23 (DR23) dye using photo-Fenton process in aqueous solution. The Fe3O4 nanoparticles were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FT-IR). Design of experiments (DOE) based on Taguchi approach was used. Analysis the response of each experiment was based Signal to Noise (S/N) ratio was calculated. The effective parameters for the degradation of dye were determined and optimized using Taguchi (L-9 (3(4))) orthogonal array experimental design method with four factors having three levels for each factor. The Taguchi approach showed that pH 3 (level 1), catalyst amount = 25 mg/L (level 2), H2O2 concentration = 15 ppm (level 3), and temperature = 35 degrees C (level 3) was optimum conditions for this process. The factor most influencing the process was determined using analysis of variance (ANOVA) method. The most significant factor in this process was pH. The interaction between pH x catalyst amount was the most influencing interaction. The percent (p (%)) of each factor on the degradation of dye was found to be in the following the order: pH (50.306%), catalyst amount (6.887%), H2O2 concentration (39.272%), and temperature (3.456%). The percentage contribution of factors in this process was found to be in the following the order: pH (0.332), catalyst amount (0.101), H2O2 concentration (0.291), and temperature (0.082). So first order reaction with k = 0.0472 min(-1) was observed for the photocatalytic degradation reaction.

We report here the stereoselective bioreduction of -nitro ketones catalyzed by ketoreductases (KREDs) with publicly known sequences. YGL039w and RasADH/SyADH were able to reduce 23 class I substrates (1-aryl-2-nitro-1-ethanone (1)) and ten class II substrates (1-aryloxy-3-nitro-2-propanone (4)) to furnish both enantiomers of the corresponding -nitro alcohols, with good-to-excellent conversions (up to >99%) and enantioselectivities (up to >99% ee) being achieved in most cases. To the best of our knowledge, KRED-mediated reduction of class II -nitro ketones (1-aryloxy-3-nitro-2-propanone (4)) is unprecedented. Select -nitro alcohols, including the synthetic intermediates of bioactive molecules (R)-tembamide, (S)-tembamide, (S)-moprolol, (S)-toliprolol and (S)-propanolol, were stereoselectively synthesized in preparative scale with 42% to 90% isolated yields, showcasing the practical potential of our developed system in organic synthesis. Finally, the advantage of using KREDs with known sequence was demonstrated by whole-cell catalysis, in which -nitro alcohol (R)-2k, the key synthetic intermediate of hypoglycemic natural product (R)-tembamide, was produced in a space-time yield of 178 g L-1 d(-1) as well as 95% ee by employing the whole cells of a recombinant E. coli strain coexpressing RasADH and glucose dehydrogenase as the biocatalyst.

Copper (II) immobilized on L-arginine-beta-cyclodextrin-functionalized magnetite nanoparticles (nano-Fe3O4@L-arginine-CD-Cu(II)) were successfully synthesized and fully characterized using FT-IR, XRD, SEM, EDX, ICP, TGA and VSM techniques. The catalytic activity of these magnetically retrievable nanoparticles was evaluated in the direct oxidation of primary alcohols and benzyl halides to acids in neat conditions that was observed to proceed well and products were obtained in good yields. Background:Licensed therapies for nonalcoholic fatty liver disease (NAFLD) do not yet exist, but clinical trials are testing treatment options. Inclusion criteria often require liver biopsy showing fibrosis (F2/3) or cirrhosis (F4) and nonalcoholic steatohepatitis (NASH). However, histological criteria pose a serious obstacle for recruitment. Aims:Characterize the relevance of liver biopsies in the selection of patients with NAFLD. Methods:Patients between 2013 and 2018 with the ICD-10 code K76.0 were analyzed. Fibrosis was defined by the NASH clinical research network (CRN) fibrosis staging system, NASH by a NAFLD activity score (NAS) >= 4. Predictive factors were determined by logistic regression. Results:Liver biopsy was performed in 87/638 (13.6%) patients (49% female, age 52.5 +/- 14.0, BMI 30.4 +/- 5.9 kg/m(2)). Fibrosis stage F0/F1/F2/F3/F4 was observed inN = 7/47/7/17/9, an NAS >= 4 inN = 27. Fibrosis stage F2/F3 and F4 along with NAS >= 4 was found in 1.7% and 0.5% of cases. Liver stiffness measurement, LSM (OR 2.3 per doubling of value; CI 1.3-4.4,p = .005) and FIB-4 (OR 2.3 per doubling of value; CI 1.2-4.4,p = .012) were significant predictors for fibrosis >= F2. Predictive factors for NASH were not identified. Conclusion:The biopsy rate in NAFLD patients is low and fibrosis >= F2 along with NAS >= 4 only present in a few cases. Transient elastography and FIB-4 are useful to select patients at risk for fibrosis for liver biopsy.

Cocrystallization of the widely available 4-dimethylaminopyridine, with a series of carboxylic acids gave nine molecular adducts 4-dimethylaminopyridinium trifluoroacetate (1) , 4-dimethylaminopyridinium glycolate glycolic acid (2) , 4-dimethylaminopyridinium 2-furoate dihydrate (3) , tri(4-dimethylaminopyridinium) tri(o-chlorobenzoate) tetrahydrate (4) , 4-dimethylaminopyridinium 4-bromobenzoate 4-bromobenzoic acid (5) , 4-dimethylaminopyridinium 4-formylbenzoate 4-formylbenzoic acid (6) , 4-dimethylaminopyridinium 2,4-dichlorobenzoate (7) , 4-dimethylaminopyridinium 2-aminobenzoate 2-aminobenzoic acid (8) and bis(4-dimethylaminopyridinium) 5-nitroisophthalate 5-nitroisophthalic acid methanol solvate (9) . The nine adducts have been characterised by X-ray diffraction, IR and elemental analysis, the melting points were also reported. Their structural and supramolecular aspects are fully analyzed. The result reveals that among the nine investigated crystals only the ring Ns in the 4-dimethylaminopyridine moieties are protonated when the carboxylic acids are deprotonated. The crystal packing is interpreted by the strong charge-assisted N-H center dot center dot center dot O hydrogen bond between the NH+ and the deprotonated acidic groups. Except the N-H center dot center dot center dot O hydrogen bonds, the charge assisted or neutral O-H center dot center dot center dot O hydrogen bonds were also found at 2-6, 8 and 9 . The COOH center dot center dot center dot COO- synthon has been at 2, 5-6 and 8-9. Further analysis of the crystal packing indicated that a different family of additional CH-O/CH2-O/CH3-O, F-F, Br-Br, CH-Cl, CH3-Cl, CH-pi/CH3-pi, C-H center dot center dot center dot C-pi, C-pi, Cl-O, O-C, O-pi and pi-pi contacts contribute to the stabilization and expansion of the total high-dimensional (2D-3D) framework structures. Aryl ketones are ubiquitous structural motifs in organic synthesis. Nonetheless, the traditional methods for their generation are often costly and not environment-friendly. Herein we report a novel and efficient route for the preparation of aryl ketones by photocatalysis at mild conditions. Porous C3N4 nanorods decorated with spatially separated Ag and Co3O4 nanoparticles on the interior and exterior surface of C(3)N(4)( )hollow sphere (Co3O4/Ag@C3N4) fabricated by a facile method of supramolecular self-assembly. Using this porous material as photocatalyst and O-2 as oxidant, aryl ketones could efficiently generate (with a yield up to 75.7% and selectivity to 97%) via toluene and halobenzene under visible-light irradiation at room temperature. The present study not only provides an inexpensive strategy for aryl ketones generation but also an effective tactic for activation of sp(3 )C-H bonds under mild conditions. (C) 2019 Elsevier Ltd. Protein kinases play a crucial role in many cell signaling processes, making them one of the most important families of drug targets. In this context, fragment-based drug design strategies have been successfully applied to develop novel kinase inhibitors. These strategies usually follow a knowledge-driven approach to optimize a focused set of fragments to a potent kinase inhibitor. Alternatively, KinFragLib explores and extends the chemical space of kinase inhibitors using data-driven fragmentation and recombination. The method builds on available structural kinome data from the KLIFS database for over 2500 kinase DFG-in structures cocrystallized with noncovalent kinase ligands. The computational fragmentation method splits the ligands into fragments with respect to their 3D proximity to six predefined functionally relevant subpocket centers. The resulting fragment library consists of six subpocket pools with over 7000 fragments, available at https://github.com/volkamerlab/KinFragLib. KinFragLib offers two main applications: on the one hand, in-depth analyses of the chemical space of known kinase inhibitors, subpocket characteristics, and connections, and on the other hand, subpocket-informed recombination of fragments to generate potential novel inhibitors. The latter showed that recombining only a subset of 624 representative fragments generated 6.7 million molecules. This combinatorial library contains, besides some known kinase inhibitors, more than 99% novel chemical matter compared to ChEMBL and 63% molecules compliant with Lipinski’s rule of five.
