Electrochemical performance and enhanced nitrate removal of homogeneous polysulfone-based anion exchange membrane applied in membrane capacitive deionization cell

This paper aims to investigate different strategies toward the enhancement of nitrate removal in a membrane capacitive deionization cell using different membrane functionalizing agents such as trimethylamine, triethylamine, and N,N,N’,N’-tetramethyl-1,6-hexanediamine as well as a double-layer, novel homogeneous polysulfone-based anion exchange membrane. The prepared membranes exhibited nitrate versus chloride transport number ratio in the range of 0.96-1.52. The salt adsorption capacity for the NaNO3 feed was about 1.4-fold compared to that of using NaCl in case of triethylamine functionalized membrane under individual feeding solutions of 30 mL NaCl or NaNO3 during 30 min, and adsorption/desorption voltages of 1.6/0.0 V process conditions. At the same time, this parameter was about 1.2-fold weaker for the cell with a double-layer membrane. Based on the experiments, it can be inferred that the affinity toward nitrate could be promoted as the chain length of the alkyl group increases (i.e., increasing hydrophobicity of the groups). However, due to decrement in water uptake, the overall electrochemical performance of the membranes tended to weaken as the affinity toward nitrate increased. Disulfiram-copper activates chloride currents and induces apoptosis with tyrosine kinase in prostate cancer cells

Aim: To elucidates the mechanism that disulfiram/copper complex (DSF/Cu) treatment activates chloride channels and induces apoptosis in prostate cancer cells. Methods: Cellular membrane currents were measured by membrane clamp technique; western blot to detect protein expression; flow cytometry to detect apoptosis; immunofluorescence to detect target protein co-localization, and further validated by a combination of protein-protein interaction and mock protein molecular docking techniques. Results: DSF/Cu activated chloride channels and induced apoptosis in LNCaP (a type of androgen-dependent prostate cancer cells) cells. The chloride currents activated by DSF/Cu were significantly reduced after knockdown of CLC3 with siRNA. In addition, DSF/Cu-activated chloride currents were reduced to background current levels after perfusion with genistein, a highly specific tyrosine kinase inhibitor. Conversely, DSF/Cu failed to activate chloride currents in LNCaP cells after 30 minutes of pre-incubation with genistein. When genistein was removed, and DSF/Cu was added, the activated currents were small and unstable, and gradually decreased. Immunofluorescence in LNCaP cells also showed co-localization of the CLC3 protein with tyrosine kinase 2 beta (PTK2B). Conclusion: DSF/Cu can activate chloride channels and induce apoptosis in LNCaP cells with the involvement of tyrosine kinase. These results provide new insights into the target therapy of prostate cancer.

How Pure and Hydrated Reline Deep Eutectic Solvents Affect the Conformation and Stability of Lysozyme: Insights from Atomistic Molecular Dynamics Simulations

Factors governing the stability and activity of proteins and enzymes in nonaqueous solvents have just been started to be explored. Because of their benign and economically viable nature, deep eutectic solvents (DESs) are being seen as an alternative media in many biotransformation processes. The present study exploits the changes in the conformation and stability of hen egg white lysozyme (HEWL) in the presence of reline (a eutectic mixture of choline chloride and urea) and reline/water mixtures using atomistic molecular dynamics simulations. The lysozyme structure was found to be partially folded in both reline and reline/water mixtures. Root-mean-square deviation (RMSD) of the positions of Ca atoms of lysozyme indicate that 50/50 reline/water solvent induces more destabilization in the conformation of HEWL than that by pure reline and 75/25 reline/water mixture. From the root-mean-square fluctuation (RMSF) analysis, it is found that the lysozyme active site (Glu35-Asp52) is quite stable in the presence of pure reline but it is least stable in the presence of 50/50 reline/water mixture. Our results show that the secondary structure of the lysozyme is significantly affected in the presence of reline. Our further analysis reveals that the hydrogen bonding interaction between HEWL-[Ch](+) dominates over HEWL-urea and HEWL-Cl- in pure reline than in reline/water mixtures.

Effectiveness of a formulation containing peptides and vitamin C in treating signs of facial ageing: three clinical studies

Anesthetic potential of the essential oils of Aloysia triphylla, Lippia sidoides and Mentha piperita for Colossoma macropomum

The aim of this study was to evaluate the anesthetic potential of the essential oils of Aloysia triphylla (EOAT), Lippia sidoides (EOLS) and Mentha piperita (EOMP) in relation to juvenile tambaqui (Colossoma macropomum), based the swimming behavior of these fish and possible histopathological damage to the gills. To induce anesthesia, juvenile tambaqui (130.55 +/- 22.4 g; 16.8 +/- 0.95 cm) were individually exposed to different concentrations of the essential oils (EOAT: 5, 10, 20, 40, 60, 80 and 150 mg L-1; EOLS: 2, 5, 10, 20, 30, 40 and 50 mg L-1; EOMP: 5, 10, 20, 40, 60, 70 and 90 mg L-1), for exposure periods of between four minutes and six hours. The three essential oils evaluated presented anesthetic properties. The concentration intervals over which the fish reached all the stages of anesthesia (1, 2, 3a, 3b and 4) were between 40 and 150 mg L-1 for EOAT; 20 and 50 mg L-1 for EOLS; and 40 and 90 mg L-1 for EOMP. The times taken to reach the anesthesia stages were inversely proportional to the increases in the concentration of the essential oils. The histopathological damage to the gills of the tambaqui that resulted from using essential oil concentrations that were defined as anesthetic (150 mg L-1 EOAT, 30 mg L-1 EOLS and 90 mg L-1 EOMP) were also evaluated. The main damage recorded was hypertrophy and hyperplasia of the lamellar epithelium, lamellar fusion and proliferation of chloride cells. According to the histopathological alteration index (HAI), there were no irreversible damage (such as fibrosis and necrosis) to the gills of the tambaqui. However, while EOLS and EOMP caused mild damages in the gills, the EOAT generated more severe damages. Abattoirs are positioned close to freshwaters in most developing countries where untreated wastewaters are discharged into with potential risk to aquatic life such as fish and ultimately human health. We assessed physicochemical parameters, antibiotics (oxytetracycline and diclofenac) and algal load of effluent collected from a major abattoir in Nigeria. Furthermore, liver function enzymes, genotoxic and haemotoxic effects of subacute concentration (10% of 96 hLC(50) value) of the wastewater were evaluated over a period of 28 d in Clarias gariepinus (The African Sharptooth Catfish). The 96 hLC(50) value of the abattoir wastewater against C. gariepinus was 154.14 mL/L (15.4%). Nitrates, phosphates, sulphates, chloride, ammonia, TDS, TSS, BOD5 and heavy metals (Fe and Pb) in the wastewater were above permissible limits while diclofenac and oxytetracycline were below detection limit (BDL). Microalgae in the wastewater were mostly Bacilliariophyta (Navicula spp.) (45.64%) and euglenoids (Euglena and Phacus spp.) (49.48%). Liver function enzymes (LDH, AST, ALT) level were higher in exposed fishes except for ALP which was lower at day 28 compared to control levels. Erythrocytic genotoxic indices (nuclear abnormalities) were significantly higher (p < 0.05) in the exposed fishes particularly at day 28 compared to control. Haematologic indices level such as WBC, MCV MCH, MCHC increased significantly (p < 0.05) while lymphocytes, HGB, RBC, HCT levels decreased significantly (p < 0.05) in the exposed C. gariepinus by day 28 compared to control. These results demonstrate potential adverse effects posed to aquatic fish species in the Ogun River by the discharge of the abattoir effluent. The microalgae species identified in the effluent may be explored for pre-treatment of the effluent before discharge in order to prevent eutrophication and increased pollutant load in the River. Uranium(VI) toxicity in tobacco BY-2 cell suspension culture – A physiological study

For the first time, the physiological and cellular responses of Nicotiana tabacum (BY-2) cells to uranium (U) as an abiotic stressor were studied using a multi-analytic approach that combined biochemical analysis, thermodynamic modeling and spectroscopic studies. The goal of this investigation was to determine the U threshold toxicity in tobacco BY-2 cells, the influence of U on the homeostasis of micro-macro essential nutrients, as well as the effect of Fe starvation on U bioassociation in cultured BY-2 cells. Our findings demonstrated that U interferes with the homeostasis of essential elements. The interaction of U with BY-2 cells confirmed both timeand concentration-dependent kinetics. Under Fe deficiency, a reduced level of U was detected in the cells compared to Fe-sufficient conditions. Interestingly, blocking the Ca channels with gadolinium chloride caused a decrease in U concentration in the BY-2 cells. Spectroscopic studies evidenced changes in the U speciation in the culture media with increasing exposure time under both Fe-sufficient and deficient conditions, leading us to conclude that different stress response reactions are related to Fe metabolism. Moreover, it is suggested that U toxicity in BY-2 cells is highly dependent on the existence of other micro-macro elements as shown by negative synergistic effects of U and Fe on cell viability.
