Penicillium raistrickii-mediated 15 alpha-hydroxylation of D-ethylgonendione is a key step for industrial production of gestodene. However, the process efficiency is limited due to the poor solubility of the steroid in aqueous water system. Deep eutectic solvents (DESs) are promising solvents applied widely in biocatalysis. Here we systematically investigated the biotransformation performance of P.raistrickii in 14 different DESs containing system. DESs with different HBD exhibited different effects on the biotransformation, and no production was found in organic acid based DESs, whereas improved biotransformation efficiency in the presence of DESs with alcohol was observed. DESs also showed synergetic effects on steroid hydroxylation when compared with that in aqueous solutions of the deep eutectic solvent ([ChCl][Gly], 1:2), its components (glycol and choline chloride), and the mixture of them, respectively. Of all the DES tested, [ChCl][Gly] was successfully applied in steroid hydroxylation, and the substrate bioconversion (72 h) enhanced to 82% in the system containing 4% DES, 2 g/L substrate and 17.5 gdw/L mycelium, which is much higher than that without DESs (33%). DES with alcohol as HBD especially [ChCl][Gly] showed better biocompability toward P.raistrickii, and their beneficial effect on mass transfer of the substrate was also observed. Results obtained in this study suggested that [ChCl][Gly] is the most potential in D-ethylgonendione hydroxylation mediated by P. raistrickii, broadening their applications in steroid production.

Ginger rhizomes have been reportedly used in folk medicine for the management of various ailments. This study, therefore, investigates the ameliorative effect of the ethanolic extract of ginger (Zingiber officinale) rhizomes against DNA damage in rats induced with different carcinogens. Fifteen Wistar rats grouped into 3 of 5 rats per group were used for the study. The first set of blood samples was first collected before the animals were orally treated with heavy metals. After 14 days of induction, the second set of blood was collected. The third phase of blood collection was done after administering an ethanolic extract of Z. officinale for 14 days. The UV wavelength absorption spectrum and conventional PCR analysis were carried out on DNA extracts of all the animals. Cluster analysis of optical density (OD) and PCR data were carried out as well as genomic instability, similarity, and diversity using the best 3 Random Amplified Polymorphic DNA (RAPD) primers. The PCR -DNA concentration analysis showed the Z. officinale extract’s ameliorative effect against lead acetate, cadmium chloride, and arsenic trioxide- induced DNA damage with a significant (p < 0.05) reduction in DNA concentration of the treated rats when compared with induced rats. The cluster analysis of optical density values revealed close similarity between the control animals' DNA, a slight similarity with treated animals' DNA, and a significant difference with the induced animal DNA. These results indicated the ameliorative properties of Z. officinale against these heavy metals induced DNA damage in rats. (C) 2020 by the authors. Markov processes provide a popular approach to construct low-dimensional dynamical models of a complex biomolecular system. By partitioning the conformational space into metastable states, protein dynamics can be approximated in terms of memory-less jumps between these states, resulting in a Markov state model (MSM). If these conditions are in conflict (i.e., tau (M) > tau (S)), one may opt for a short time step delta t = tau (S) and try to account for the residual non-Markovianity of the data by optimizing the transition matrix or the Langevin fields such that the resulting model best reproduces the observables of interest. In this work, rescaling the friction tensor of the dLE based on short-time information in order to obtain the correct long-time behavior of the system is suggested. Adopting various model problems of increasing complexity, including a double-well system, the dissociation of solvated sodium chloride, and the functional dynamics of T4 lysozyme, the virtues and shortcomings of the rescaled dLE are discussed and compared to the corresponding MSMs.

The human glycine transporter 1 (GlyT1) regulates glycine-mediated neuronal excitation and inhibition through the sodium- and chloride-dependent reuptake of glycine(1-3). Inhibition of GlyT1 prolongs neurotransmitter signalling, and has long been a key strategy in the development of therapies for a broad range of disorders of the central nervous system, including schizophrenia and cognitive impairments(4). Here, using a synthetic single-domain antibody (sybody) and serial synchrotron crystallography, we have determined the structure of GlyT1 in complex with a benzoylpiperazine chemotype inhibitor at 3.4 angstrom resolution. We find that the inhibitor locks GlyT1 in an inward-open conformation and binds at the intracellular gate of the release pathway, overlapping with the glycine-release site. The inhibitor is likely to reach GlyT1 from the cytoplasmic leaflet of the plasma membrane. Our results define the mechanism of inhibition and enable the rational design of new, clinically efficacious GlyT1 inhibitors.

Ethnopharmacological relevance: Launaea acanthodes (Boiss.) O. Kuntze is native to semiarid regions of central Iran, traditionally used in the treatment of numerous disorders including diabetes. Aim of the study: The current study aimed to explore hypoglycemic activity of Launaea acanthodes extract in streptozotocin-induced diabetic rats. Furthermore, gene expression study was carried out to examine expression levels of key glucose metabolism-related genes. Methods: For in vitro study, Folin-Ciocalteus, DPPH and aluminum chloride colorimetric assays were used to determine the total phenolic content, antioxidant capacity and total flavonoid content of extracts, respectively. For in vivo study, streptozotocin-induced diabetic Wistar rats were orally administered with metformin (50 mg/ kg) and various doses of extracts (100, 200 and 400 mg/kg body weight) for 28 days. Fasting blood glucose, body weight, food and water intake were assessed during the course of treatment. At the end of the intervention, oral glucose tolerance test (OGTT), lipid profile and glycated hemoglobin (HbA1c) were evaluated. Furthermore, functional liver enzymes, oxidative stress markers and histopathology of pancreas were examined. Lastly, quantitative real time polymerase chain reaction (qRT-PCR) was applied to explore the mRNA levels of genes relevant to glucose metabolism in the pancreas and liver tissues of diabetic rats. Results: Based on the in vitro results, the hydroalcoholic extract revealed potential radical scavenging activity and contained highest amount of phenolic and flavonoid. The in vivo results demonstrated that the extract lowered fasting blood glucose level, increased the body weight, restored the alterations in the levels of water and food intake, attenuated HbA1c, improved lipid profile and ameliorated the OGTT in diabetic rats. The extract administration alleviated the histopathological changes in the pancreas, suppressed malondialdehyde (MDA) level and further restored attenuated levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), glutathione peroxidase (GPx) and superoxide dismutase (SOD) in diabetic rats. Analysis of real time PCR data showed that extract administration reversed the expression levels of hepatic glucokinase (GK), phosphenolpyruvate carboxykinase (PEPCK) and glucose-6-phosphatase (G6Pase). Meanwhile, the extract upregulated the expression level of glucose transporter-2 (GLUT-2) and pancreatic-duodenal homeobox (PDX-1) in diabetic rats. Conclusion: Collectively, the results demonstrate that Launaea acanthodes hydroalcoholic extract exerts hypoglycemic effect possibly via regulating key enzymes of glucose metabolism and ameliorating pancreatic dysfunction through its antioxidant properties.

Persistent anion binding in a wide range of solution environments is a key challenge that continues to motivate and demand new strategies in synthetic receptor design. Though strong binding in low-polarity solvents has become routine, our ability to maintain high affinities in high-polarity solvents has not yet reached the standard set by nature. Anions are bound and transported regularly in aqueous environments by proteins that use secondary and tertiary structure to isolate anion binding sites from water. Inspired by this principle of solvent exclusion, we created a sequence-defined foldameric capsule whose global minimum conformation displays a helical folded state and is preorganized for 1:1 anion complexation. The high stability of the folded geometry and its ability to exclude solvent were supported by solid-state and solution phase studies. This capsule then withstood a 4-fold increase in solvent dielectric constant (epsilon(r)) from dichloromethane (9) to acetonitrile (36) while maintaining a high and solvent-independent affinity of 10(5) M-1; Delta G similar to 28 kJ mol(-1). This behavior is unusual. More typical of solvent-dependent behavior, Cl- affinities were seen to plummet in control compounds, such as aryl-triazole macrocycles and pentads, with their solvent-exposed binding cavities susceptible to dielectric screening. Finally, dimethyl sulfoxide denatures the foldamer by putative solvent binding, which then lowers the foldamer’s Cl- affinity to normal levels. The design of this capsule demonstrates a new prototype for the development of potent receptors that can operate in polar solvents and has the potential to help manage hydrophilic anions present in the hydrosphere and biosphere.

Biocides such as quaternary ammonium compounds (QACs) are potentially important contributors towards bacterial antimicrobial resistance development, however, their contributions are unclear due to a lack of internationally recognized biocide testing standards. Methods to detect QAC tolerance are limited to laborious traditional antimicrobial susceptibility testing (AST) methods. Here, we developed a rapid fluorescent dye-based membrane impermeant assay (RFDMIA) to discriminate QAC susceptibility among Gram-negative Enterobacterales and Pseudomonadales species. RFDMIA uses a membrane impermeant fluorescent dye, propidium iodide, in a 30-min 96-well fluorescent microplate-based assay where cell suspensions are exposed to increasing QAC concentrations. Our results demonstrate that RFDMIA can discriminate between QAC-susceptible and QAC-adapted Escherichia coli tolerant phenotypes and predict benzalkonium and cetrimide tolerance in all species tested except for intrinsically fluorescent Pseudomonas aeruginosa. RFDMIA identified a close association to minimum inhibitory concentration values determined by broth microdilution AST and increasing fluorescent dye emission values. RFDMIA emission values and scanning electron microscopy results also suggest that CET-adapted E. coli isolates have a CET dependence, where cells require sub-inhibitory CET concentrations to maintain bacilliform cell integrity. Overall, this study generates a new, rapid, sensitive fluorescent assay capable of detecting QAC-susceptible Gram-negative bacteria phenotypes and cell membrane perturbations.

Silver nanowires were successfully prepared by polyol method using silver nitrate as silver source, ethylene glycol as solvent and reducing agent, and ferric chloride as nucleating agent and polyvinylpyrrolidone (PVP) as structure directing agent and protective agent. The obtained silver nanowires were mixed with Kraft pulp fibers and bacterial cellulose to prepare conductive paper via simple filtration process. The results indicated that the size of silver nanowires can be regulated by the dosage of ferric chloride used. At optimal dosage of FeCl3, the prepared silver nanowires can reach 20 mu m length, 100 nm width, and 200 of width/diameter ratio. The adding of bacterial cellulose in preparing the conductive paper greatly enhanced the retention ratio of silver nanowires as well as the stress and strain of the conductive paper. The stress and strain of the paper with bacterial cellulose were increased by 60% and 7%, respectively, compared with that of the paper without bacterial cellulose. The maximum retention ratio and the highest recovery ratio of silver nanowires in conductive paper reached 94% and 75%, respectively. The resistance of as-prepared conductive paper was only 0.9 omega. In conductive paper, the silver nanowires attached to the paper-based materials with cellulose fibers as the substrate, which achieved the electrical conductivity of paper-based materials. This conductive paper can be applied as antistatic, antibacterial packaging and conductive material.

This study presents the evaluation of an air conditioning system with enthalpy recovery for space heating and air humidification. Desiccant assisted air conditioning is potentially beneficial against other air conditioning processes in terms of air humidification, counteracting dry indoor air conditions during winter. Furthermore, the overall process can be operated efficiently relying on renewable energy sources. The objective of this work is to investigate a geothermal and desiccant assisted system for different system configurations experimentally and numerically during winter operation. The experimental investigation focuses on differences in system performance for two desiccant materials, lithium chloride and silica gel. Experimental results show that moisture recovery relying on a desiccant wheel with silica gel is beneficial against lithium chloride. An average moisture recovery efficiency of 0.83 was achieved for silica gel. Thermal comfort was maintained on a high level throughout the winter seasons investigated. Based on component and system simulation models, system performance is shown for different boundary conditions during winter requiring space heating and humidification to ensure comfortable indoor air conditions. The simulation results show a reduction in primary energy demand for heat supply by up to 54% by using a high efficient ground-coupled heat pump without backup system. (C) 2020 The Author(s).
