Apoptosis-antagonizing transcription factor is involved in rat post-traumatic epilepsy pathogenesis

The present study aimed to explore the pathogenesis behind post-traumatic epilepsy (PTE). In the present study, a chloride ferric injection-induced rat PTE model was established. The expression levels of apoptosis-antagonizing transcription factor (AATF), cleaved caspase-3, p53, Bcl-2 and Bax were measured by western blotting or immunofluorescence staining (IF). The expression of AATF in vivo was downregulated by microinjection of lentiviral-mediated short-hairpin RNA. Compared with control and sham groups, at day 5 after PTE, neuron apoptosis was significantly increased and the expression levels of AATF, p53, cleaved caspase-3 and Bax were significantly upregulated. In addition, IF revealed co-localization of AATF and cleaved caspase-3 in the cortex. Additionally, AATF was expressed mainly in neurons and astrocytes. Following AATF inhibition, the expression levels of p53 and cleaved caspase-3 were significantly reduced as compared with the control group. Taken together, these findings suggested that following PTE, AATF is involved in neuronal apoptosis and may serve as a potential target for its alleviation.

Investigation of PolyVinyl Chloride Plastisol Tissue-Mimicking Phantoms for MR- and Ultrasound-Elastography

Objective: Realistic tissue-mimicking phantoms are essential for the development, the investigation and the calibration of medical imaging techniques and protocols. Because it requires taking both mechanical and imaging properties into account, the development of robust, calibrated phantoms is a major challenge in elastography. Soft polyvinyl chloride gels in a liquid plasticizer (plastisol or PVCP) have been proposed as soft tissue-mimicking phantoms (TMP) for elasticity imaging. PVCP phantoms are relatively low-cost and can be easily stored over long time periods without any specific requirements. In this work, the preparation of a PVCP gel phantom for both MR and ultrasound-elastography is proposed and its acoustic, NMR and mechanical properties are studied. Materials and methods: The acoustic and magnetic resonance imaging properties of PVCP are measured for different mass ratios between ultrasound speckle particles and PVCP solution, and between resin and plasticizer. The linear mechanical properties of plastisol samples are then investigated over time using not only indentation tests, but also MR and ultrasound-elastography clinical protocols. These properties are compared to typical values reported for biological soft tissues and to the values found in the literature for PVCP gels. Results and conclusions: After a period of two weeks, the mechanical properties of the plastisol samples measured with indentation testing are stable for at least the following 4 weeks (end of follow-up period 43 days after gelation-fusion). Neither the mechanical nor the NMR properties of plastisol gels were found to be affected by the addition of cellulose as acoustic speckle. Mechanical properties of the proposed gels were successfully characterized by clinical, commercially-available MR Elastography and sonoelastography protocols. PVCP with a mass ratio of ultrasound speckle particles of 0.6%-0.8% and a mass ratio between resin and plasticizer between 50 and 70% appears as a good TMP candidate that can be used with both MR and ultrasound-based elastography methods.

Nanoencapsulation of food bioactives in supramolecular assemblies based on cyclodextrins and surfactant

The bioavailability of poorly soluble food ingredients can be achieved by their encapsulation into the 3D macrocyclic structure or self-assembled surfactant micelles. In this study, the mixed self-assembly of cyclodextrins (CDs) and cationic surfactant, 1-hexadecyl-4-(2-(1-methylpyridin-1-ium-4-yl)vinyl)pyridin-1-ium chloride, was studied by a set of physicochemical methods. It was shown using tensiometry and conductometry that the presence of CDs in the micellar medium of this surfactant increases the critical aggregation concentration. Moreover, NMR spectroscopy and spectrophotometry revealed the interaction between the alkyl tail of surfactant and the internal cavity of CDs. It is interesting to note that in the case of gamma-CD and HP-beta-CD there was additional binding by the vinyl group of surfactant. The observed patterns of aggregation behavior were confirmed by the method of solubilization of hydrophobic substances. Spectrophotometric monitoring of the solubilization of hydrophobic flavonoids (rutin and quercetin) demonstrated that mixed assemblies enriched with surfactant are capable of binding the flavonoid, while they show poor binding ability towards flavonol compound in the presence of a small amount of surfactant.. Variation of the component ratio in the supramolecular CD-surfactant system also allows adjusting the biological properties. Systems with an excess of CD are less toxic to the cells of test microorganisms, herewith, an increase of the surfactant content in a mixture with CD leads to an increase in both antimicrobial and hemolytic activities. Chemically Durable Poly(phenylene-co-arylene ether) Multiblock Copolymer-Based Anion Exchange Membranes with Different Hydrophobic Moieties for Application in Fuel Cells

Quaternized multiblock poly(phenylene-co-arylene ether)s (QPPs) are synthesized from a pre-aminated monomer and a series of chlorine-terminated oligo(arylene-ethers) via a nickel coupling reaction. Their properties are investigated as anion exchange membranes with the goal of obtaining membranes with enhanced performance and durability for alkaline membrane fuel cell applications. Three different oligo(arylene ethers) were synthesized and introduced in the hydrophobic block of the polymer backbone to study its effect on the membrane properties. The chloride ion conductivity of the QPP membranes was significantly less dependent on the humidity compared to that of the commercial Fumatech membrane due to the well-defined hydrophilic-hydrophobic phase separation, as confirmed by TEM. An accelerated alkaline stability test indicated that the QPPs were more stable than the commercial Fumatech membrane up to 1700 h. The QPP-6F membrane shows an alkaline membrane fuel cell single-cell performance with a maximum power density of 534 mW cm(-2) at 70 degrees C.

The kinetics of the initial Py-/ substitution reaction are shown to follow a modified form of the two-term rate law typical of square planar complexes. The observation of a slow, but spontaneous, conversion of Au-III species to Au-I species has been explained by estimating the standard reduction potential for the pyridine/pyridine-N-oxide redox couple and demonstrating that it can drive a galvanic reaction with the AuPy/Cl-2(q)(OH)(2-q)(+)/AuPy2/+ redox couple. The previously reported tendency of this system to form highly anisotropic gold nanoparticles on conductive metal oxide electrodes is explained by a thermodynamic analysis of the disproportionation reaction of AuPy2/+ (C) 2020 Elsevier Ltd. Activation of peroxymonosulfate by calcined electroplating sludge for ofloxacin degradation

Developing cost-effective metal/metal oxides for peroxymonosulfate (PMS) activation remains a key issue in the sulfate radical based advanced oxidation process. In this work, electroplating sludge (ES), a transition metal-rich byproduct, was anaerobic calcined and characterized. Then, calcined electroplating sludge (CES) was applied as PMS activator for degradation of ofloxacin (OFL) and CES/PMS system exhibited a nearly 90% of OFL removal in 60 min. In addition, effect of CES, PMS, the initial pH and water constituents (chloride, bicarbonate, natural organic matter (NOM) and water backgrounds) on OFL degradation were systematically studied. Moreover, radical quenching tests and electron spin-resonance spectroscopy studies manifested that both SO4 center dot- and HO center dot were the ruling reactive oxygen species. X-ray photoelectron spectroscopy results of the fresh and used CES demonstrated that the PMS activation mainly occur in the transformation from Fe3+ (Cu2+) to Fe2+ (Cu+). Furthermore, liquid chromatography coupled with ion trap time-of-flight mass spectrometry was used to illustrate the possible degradation pathway of OFL. Moreover, CES showed excellent stability and reusability during reaction. Enhanced removal of aqueous phenol with polyimide ultrafiltration membranes embedded with deep eutectic solvent-coated nanosilica

Novel composite hydrophilic ultrafiltration (UF) membranes, comprising a polyimide (PI) matrix embedded with silica-based nanofillers are established for the enhanced vacuum-filtration removal of small polar organics such as phenol from water. Spherical silica nanoparticles are first prepared by the surfactant-templated sol-gel approach, then coated with choline chloride-ethylene glycol deep eutectic solvent (DES) by post-impregnation. The modified silica nanofillers are introduced into the PI matrix through the casting solution, which is then used to develop the composite UF membranes by phase inversion. The spectroscopy and high-resolution electron microscopy measurements evidence the presence of DES on the surface of the modified silica nanofillers. The topographical atomic force microscopy studies, dynamic water contact angle, and zeta potential measurements conducted on the native polymer membrane and those embedded with bare-silica and DES-coated silica confirm the successful incorporation of nanofillers into the PI matrix. An optimum nanofiller loading of 2 wt% is found to preserve the membrane structure and give the best mechanical properties as well as for the UF performance characteristics in vacuum-filtration studies at 85 kPa. Consistently, the DES-silica modified PI UF membranes (DES@SiO2@PI) exhibit exceptional water permeate flux (300 L m(2) h(-1)) and removal efficiency (96%) for 30 mg L-1 aqueous phenol compared with the native polymer and SiO2-modified membranes. The DES-engineered composite membranes can be operated within a wide feed pH window (pH: 2-10) without displaying significant loss in their removal efficiency. The pH and zeta potential results suggest that the interaction of phenol with the optimized membranes is governed by cooperative adsorption/Lewis acid-base mechanisms via hydrogen bonding and assistive ion-exchange interactions with the ethaline coating.
